Exercise builds the scaffold of life: muscle extracellular matrix biomarker responses to physical activity, inactivity, and aging

Georgios Mavropalias et al.

Summary: This review examines the short- and long-term changes in ECM markers in biofluids and tissues, and highlights their importance in studying muscle function, growth, and structure. The effects of physical activity and disuse on ECM vary, and understanding these changes can accelerate the development of targeted physical activity treatments.

Overexpression of the Hsa21 Transcription Factor RUNX1 Modulates the Extracellular Matrix in Trisomy 21 Cells

Nunzia Mollo et al.

Summary: Down syndrome is associated with upregulation of extracellular matrix (ECM) genes in the heart. The transcription factor RUNX1, located on chromosome 21, may potentially regulate the expression of these ECM genes. Silencing of RUNX1 in trisomic fetal fibroblasts reduces the expression of ECM genes and increases cell migration capacity.

Quantitative magnetic resonance characterization of the effect of physical training on skeletal muscle of the Ts65Dn mice, a model of Down syndrome

Barbara Cisterna et al.

Summary: This study aimed to evaluate the effects of physical exercise on the hindlimbs of trisomic and euploid male mice. The results showed that one month of adapted physical exercise did not induce quantitative changes in skeletal muscle or fiber type composition in DS, but the metabolic response of skeletal muscle to exercise may be affected by trisomy. Further research is needed to clarify the mechanisms of the muscular deficit found in DS.

A computational approach to quantitatively define sarcomere dimensions and arrangement in skeletal muscle

Barbara Cisterna et al.

Summary: This study conducted a computerized analysis of myofiber and sarcomere structures, revealing significant differences in Z-line length, sarcomere length, and shape parameters in trisomic muscle compared to euploid muscle. These findings provide a quantitative basis for future research on skeletal muscle changes in pathological conditions.

Age-Related Changes in the Matrisome of the Mouse Skeletal Muscle

Francesco Demetrio Lofaro et al.

Summary: This study characterized the matrisome in the aging skeletal muscle for the first time, demonstrating significant statistical differences in several matrisome proteins between aged and adult muscle. The findings provide new insights into the mechanisms regulating myocellular behavior in aging and their contribution to age-related muscle dysfunction.

The Role of Z-disc Proteins in Myopathy and Cardiomyopathy

Kirsty Wadmore et al.

Summary: The Z-disc is crucial for maintaining the structure and signaling function of the sarcomere in muscle cells. Six key Z-disc proteins, associated with myopathies and cardiomyopathies, play a significant role in force transduction and intracellular signaling. Evaluating the pathogenic variants and minor allele frequency of these proteins in population cohorts can help in re-evaluating their pathogenicity.

Summary: This study identifies prenatal lung abnormalities in individuals with Down syndrome (DS), characterized by histopathological abnormalities, reduced cell proliferation, and dysregulated gene expression. These findings contribute to a better understanding of the pathogenesis of lung disease in DS patients.
