Quantitative mapping of mercury and selenium in mushroom fruit bodies with laser ablation-inductively coupled plasma-mass spectrometry

Simone Braeuer et al.

Summary: This work developed a novel method for quantitative mapping of Hg and Se in mushroom fruit body tissues using laser ablation coupled to inductively coupled plasma-mass spectrometry (LA-ICP-MS). The protocol for preparation of quantification standards was optimized, and a reference material was successfully prepared for method validation. The method was applied to investigate the distribution of Hg and Se in different mushroom species.

Calibration strategies for laser ablation ICP-MS in biological studies: A review

Huijie Pan et al.

Summary: Laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) has high sensitivity and sufficient spatial resolution for in-situ analysis and imaging in biological studies. However, matrix effect and elemental fractionation pose challenges for quantitative analysis. This review discusses various calibration strategies for LA-ICP-MS, including internal standardization, external calibration, isotope dilution, and machine learning-based calibration, to achieve accurate quantification in biological studies.

High-speed mapping of Hg and Se in biological tissue via laser ablation-inductively coupled plasma-mass spectrometry

Tom Van Helden et al.

Summary: This study investigates the effects of instrumental setup on the response duration of elements, such as Hg and Se, in biological tissue. By optimizing the setup, significant improvement in the response duration is achieved. However, caution should be taken when selecting instrument settings and data acquisition conditions for accurate elemental mapping to avoid smearing effects.

Toward a comprehensive study for multielemental quantitative LA-ICP MS bioimaging in soft tissues

Carine Arnaudguilhem et al.

Summary: This study developed a quantitative LA-ICP MS bioimaging methodology based on polymer film strategy and internal standardization, which can simultaneously map eight elements in biological tissue sections. Different elements exhibit varied responses to analytical conditions and internal standards, primarily dependent on First Ionization Potential. The methodology was validated for clinical human samples and showed promising results.

Determination of spatial mercury concentration by laser ablation-inductively coupled plasma mass spectrometry

Makiko Iwase et al.

Summary: A highly quantitative mercury imaging method was established utilizing LA-ICP-MS with a homogeneous mercury standard consisting of rat tissue, showing good linearity between concentration and signal intensity. The study demonstrated that LA-ICP-MS enabled quantitative imaging with the appropriate standard, with comparable results to cold vapor AAS.

Tissue localization of selenium of parental or dietary origin in rainbow trout (Oncorhynchus mykiss) fry using LA-ICP MS bioimaging

Pauline Wischhusen et al.

Summary: This study investigated the impact of different forms and levels of parental and dietary selenium supplementation on selenium content and distribution in rainbow trout fry. Results showed that the selenium levels were higher in the supplementation groups, and the form and level of selenium could influence the absorption and retention of selenium in different tissues.

Investigating the effect of species-specific calibration on the quantitative imaging of iron at mg kg-1 and selenium at μg kg-1 in tissue using laser ablation with ICP-QQQ-MS

Kharmen Billimoria et al.

Summary: This paper presents a methodology for producing matrix-matched tissue homogenates containing Fe and Se at pathophysiological levels and investigates their feasibility as calibration standards for quantitative imaging of these elements. The impact of elemental species-specific calibration on the quality of LA-ICP-MS data is evaluated, with Se calibrants having a greater effect compared to Fe. Mixing methanol with the laser-induced aerosol improved the detection limit of Se, allowing for the simultaneous detection of both elements at physiologically relevant concentrations.
