Development of an Efficient C-to-T Base-Editing System and Its Application to Cellulase Transcription Factor Precise Engineering in Thermophilic Fungus Myceliophthora thermophila

Chenyang Zhang et al.

Summary: A CRISPR/Cas-based base-editing approach has been developed to efficiently edit target genes in the thermophilic fungus Myceliophthora thermophila. The base editor, capable of introducing point mutations without causing double-strand breaks, was used to successfully inactivate genes and explore the functions of specific motifs in filamentous fungi. The findings highlight the potential of this base-editing system in synthetic biology and metabolic engineering.

MtTRC-1, a Novel Transcription Factor, Regulates Cellulase Production via Directly Modulating the Genes Expression of the Mthac-1 and Mtcbh-1 in Myceliophthora thermophila

Nan Li et al.

Summary: In this study, a novel regulator MtTRC-1 in Mycorella thermophila was identified and characterized, which regulated cellulase production by directly transcriptionally regulating the Mthac-1 and Mtcbh-1 genes. The findings demonstrate the importance of MtTRC-1 for positively regulating cellulase production in the thermophilic fungus.

Combinatorial Engineering of Transcriptional Activators in Penicillium oxalicum for Improved Production of Corn-Fiber-Degrading Enzymes

Liwei Gao et al.

Summary: Enzymatic conversion of corn fiber to fermentable sugars by Penicillium oxalicum is efficient. Engineering of transcriptional activators CIrB, XInR, and AraR leads to increased production of lignocellulolytic enzymes, with synergistic effects observed in un-pretreated corn fiber saccharification. Combinatorial engineering of all three activators generated a strain MCAX with significantly higher lignocellulolytic enzyme production and higher sugar release efficiency from corn fiber.
