Crosstalk between Yeast Cell Plasma Membrane Ergosterol Content and Cell Wall Stiffness under Acetic Acid Stress Involving Pdr18

Summary: This study investigates the impact of Pdr18 expression on yeast cell wall during adaptation to acetic acid stress. Results show that Pdr18 deletion leads to changes in transcriptional activation of cell wall biosynthetic genes and cell wall stiffness and polysaccharide content. Despite a stronger and more long-lasting adaptive response, the pdr18 Delta population has lower resistance to lyticase activity and requires a longer period for intracellular pH recovery and growth resumption. Plasma membrane ergosterol content is drastically reduced under acetic acid stress, especially in pdr18 Delta cells, suggesting a coordinated response between plasma membrane and cell wall against the detrimental effects of acetic acid.
