Digital Decoding of Single Extracellular Vesicle Phenotype Differentiates Early Malignant and Benign Lung Lesions

Summary: This study investigates the use of small extracellular vesicles (sEVs) molecular profiles in differentiating malignant and benign lung nodules via a liquid biopsy-based approach. The researchers develop a novel chip called DECODE that can analyze sEV biomarkers expression and generate digitally acquired sEV molecular profiles. The results show that DECODE can successfully separate malignant from benign lung nodules, suggesting its potential for non-invasive characterization of lung nodules found in lung cancer screening.

Three-Photon AIE Pt(II) Complexes as Cysteine-Targeting Theranostic Agents for Tumor Imaging and Chemotherapy

Summary: In this study, a series of three-photon fluorescent Pt(II) complexes targeting a tumor-associated biothiol, cysteine (Cys), were synthesized to detect Cys without interference from other intracellular proteins. The fluorescence response of Cys was significantly improved by regulating the recognition units in the probes. The synthesized complex, Lyso-Pt-CH3, showed strong three-photon absorption cross section (3PA) only in the presence of Cys, making it a potential biomarker for lysosome-related Cys in vitro. This work provides a novel strategy for the rational design of controllably activated and Cys-targeted Pt(II) anticancer prodrugs for clinical diagnosis and treatment.

Tunning Intermolecular Interaction of Peptide-Conjugated AlEgen in Nano-Confined Space for Quantitative Detection of Tumor Marker Secreted from Cells

Summary: The two steps-based MP/NPs-SLIPS sensing system was designed for ultrasensitive detection of the tumor marker MMP2 by tuning intermolecular interactions. The aggregation of AIE residual was intensified through electrostatic absorption by negatively charged NPs and confined space formed by self-assembly of NPs to PCs on SLIPS, resulting in increased fluorescent signals and improved sensitivity, reducing the limit of detection to 3.7 ng/mL.

Aggregation-induced emission luminogens reveal cell cycle-dependent telomerase activity in cancer cells

Summary: An AIEgen-based biosensing system was developed to detect telomerase in different cell cycles. The fluorescence signal of cancer cells increased gradually from G0/G1 to S phase, while cancer cells arrested at G2/M phase and normal cells exhibited negligible signal. Compared to normal tissues, malignant tumor samples demonstrated a significant turn-on fluorescence signal.

First aggregation-induced emission-active probe for species-specific detection of β-galactosidase

Summary: This study introduces a fluorescent probe HBTTPAG with aggregation-induced emission (AIE) activity for species-selective detection of beta-galactosidase. The probe shows high sensitivity, superior selectivity, and low cytotoxicity, and can discriminate between Aspergillus oryzae beta-galactosidase and Escherichia coli beta-galactosidase. It also enables visualization of beta-galactosidase in lysosomes of SKOV-3 cells and detection of beta-galactosidase activity in ovarian cancer tissues.

TEPP-46-Based AIE Fluorescent Probe for Detection and Bioimaging of PKM2 in Living Cells
