Efficient heterologous expression of cytochrome P450 enzymes in microorganisms for the biosynthesis of natural products

Summary: Natural products are structurally diverse molecules with a wide range of biological activities. Cytochrome P450 enzymes are involved in the biosynthesis of many natural products and recent research has provided new insights for their application in the biosynthesis of natural products.

Elucidation of the pathway for biosynthesis of saponin adjuvants from the soapbark tree

Summary: The Chilean soapbark tree produces QS saponins, which are important vaccine adjuvants. By sequencing the Q. saponaria genome, we identified 16 pathway enzymes and successfully produced advanced QS pathway intermediates in tobacco. We also identified the enzymes necessary for the production of QS-7, a saponin with excellent therapeutic properties. Our findings enable the production of Q. saponaria vaccine adjuvants and provide new avenues for developing immunostimulants.

High efficient production of plant flavonoids by microbial cell factories: Challenges and opportunities

Summary: Plant flavonoids are secondary metabolites with various physiological and pharmacological activities. Current production methods from plants or chemicals do not meet the requirements of green and sustainable development. Microbial synthesis of flavonoids has shown potential for large-scale production, but challenges such as metabolic flux imbalance and conflict between cell growth and production hinder commercialization of microbial cell factories. Strategies discussed in this article, combined with protein structure prediction, de novo protein design, artificial intelligence, biocatalytic retrosynthesis, and intelligent stress resistance, provide insights for efficient production of plant flavonoids and other natural products in microbial cell factories.

Metabolic engineering of Escherichia coli with electron channelling for the production of natural products

Summary: This study reports an electron channelling strategy based on the application of Photorhabdus luminescens CipB scaffold protein, which allows efficient electron transfer between eukaryotic cytochrome P450s (P450s) and P450 reductase in bacteria. Using this strategy, recombinant Escherichia coli strains producing various natural products were developed and the production titres were enhanced through optimization of the culture conditions.

A microbial supply chain for production of the anti-cancer drug vinblastine

Summary: This study demonstrates the de novo microbial biosynthesis of vindoline and catharanthine using a highly engineered yeast, and in vitro chemical coupling to vinblastine. By integrating a very long biosynthetic pathway into yeast, the researchers have provided a scalable platform for producing over 3,000 natural MIAs and numerous new analogues.

Ice2 promotes ER membrane biogenesis in yeast by inhibiting the conserved lipin phosphatase complex

Summary: Cells dynamically adjust organelle size to meet physiological demand, with organelle growth requiring coordination with lipid metabolism. The ER transmembrane protein Ice2 plays a key role in promoting ER membrane biogenesis by inhibiting the dephosphorylation and activation of Pah1, a key enzyme in lipid metabolism. Ice2 cooperates with the transcriptional regulation of lipid synthesis genes to maintain cell homeostasis during ER stress, highlighting the importance of controlling the lipin phosphatase complex in regulating ER membrane biogenesis.

De Novo Biosynthesis of the Oleanane-Type Triterpenoids of Tunicosaponins in Yeast

Summary: Tunicosaponins are natural products extracted from Psammosilene tunicoides, with representative aglycones being gypsogenin and quillaic acid. Two novel cytochrome P450-dependent monooxygenases involved in the biosynthesis of tunicosaponins were identified, and the whole biosynthesis pathway of the tunicosaponin aglycones was successfully reconstituted in yeast.
