Summary: Phosphodiesterases (PDE) type 3 and 4 promote vasoconstriction by hydrolysing cAMP. In experimental heart failure (HF), PDE3 makes PDE4 redundant in aorta, but it is not known if this occurs in resistance vessels, such as mesenteric artery. As PDE2 is increased in the failing myocardium, its possible role in the vasculature also needs to be addressed.

Summary: The effects of PDE-4 inhibitors rolipram and roflumilast on a doxorubicin-induced nephrotic syndrome model were evaluated. The results showed that early-stage rolipram treatment preserved glomerular filtration barrier function and reduced inflammation and renal fibrosis. Late-stage treatment with either rolipram or roflumilast also protected glomerular filtration barrier function and reduced renal tissue fibrosis.

Summary: Zl-n-91, a specific inhibitor of type 4 phosphodiesterase (PDE4), can suppress AML cell proliferation, block DNA replication, and induce cell death. It also inhibits the growth of subcutaneous xenograft and prolongs survival in an AML model. Bioinformatic analysis suggests that elevated mitochondrial gene signatures are inversely associated with AML patient survival, and Zl-n-91 strongly suppresses mitochondrial function. This PDE4 inhibitor also affects multiple signaling pathways, including beta-catenin activity. Activation of the Wnt/beta-catenin pathway attenuates the inhibitory effect of Zl-n-91 on AML cell proliferation and mitochondrial function. Overall, targeting PDE4 activity with specific inhibitors holds promise as a new treatment modality for AML patients.

Additionally, inhibition of H3R rescued the impairments of neurogenesis and cognitive function caused by neuroinflammation. Mechanistically, the neuroprotective effects of H3R inhibition were mediated by histamine-dependent activation of H2R and reinforcement of CREB-CBP interaction, which led to increased release of anti-inflammatory cytokines and brain-derived neurotrophic factor, as well as decreased release of pro-inflammatory cytokines.
