Integrated transcriptomic and metabolomic analysis reveals the response of pearl oyster (Pinctada fucata martensii) to long-term hypoxia

Histone deacetylase inhibitor butyrate inhibits the cellular immunity and increases the serum immunity of pearl oyster Pinctada fucata martensii

Summary: In this study, it was found that histone acetylation in pearl oyster was enhanced after lipopolysaccharide (LPS) stimulation and further increased after treatment with LPS and butyrate. Transcriptome analysis revealed that butyrate counter-regulated a significant portion of LPS-repressed genes and inhibited the up-regulation of LPS-induced genes. Gene expression analysis showed that butyrate downregulated cell proliferation and immune-related genes. The results suggest that histone deacetylase plays a role in the immune response of pearl oyster.

Integrated transcriptomic and metabolomic analysis sheds new light on adaptation of Pinctada fucata martensii to short-term hypoxic stress

Summary: Analyses of the transcriptome and metabolome in pearl oysters exposed to short-term hypoxia revealed alterations in key genes and metabolites. A total of 209 differentially expressed genes (DEGs) were detected between the control and hypoxia groups. Enrichment analysis showed that these DEGs were enriched in GO terms related to oxidation-reduction process, ECM organization, chaperone cofactor-dependent protein refolding, and ECM-receptor interaction KEGG pathway. Moreover, 28 differential metabolites (SDMs) implicated in metabolic pathways such as phenylalanine metabolism, D-amino acid metabolism, and aminoacyl-tRNA biosynthesis were identified. The findings suggest that pearl oysters experience oxidative stress and apoptosis under short-term hypoxia, and also undergo adaptive changes in antioxidant activity, immune and biomineralization processes, protein homeostasis, and cytoskeleton organization. This study helps uncover the mechanisms by which pearl oysters respond to short-term hypoxia adaptively.

Transcriptome Analysis of Crassostrea sikamea (♀)x Crassostrea gigas (♂) Hybrids Under and After Thermal Stress

Summary: Crossbreeding is an effective strategy to manage genetic decline in aquaculture. Hybrids of Crassostrea sikamea (♀) and Crassostrea gigas (♂) exhibit advantages in growth traits and high temperature adaptation. High-throughput sequencing was utilized to analyze molecular processes in the hybrids during and after thermal stress, leading to the identification of pathways enriched during thermal stress (antigen processing and presentation, NOD-like, and NF-kappa B) and recovery stage (MAPK and PPAR). The study indicates the deployment of innate immune system to protect tissues during thermal stress, with 85% of mutual DEGs involved in bidirectional regulation during recovery.

Genome-wide analysis of the hard clam mitogen-activated protein kinase kinase gene family and their transcriptional profiles under abiotic stress

The mRNA levels of certain genes in the liver and intestine were also affected by dietary lysine levels.
