Netrin-1: An emerging player in inflammatory diseases

Xueli Xia et al.

Summary: Netrin-1, a chemotactic molecule, participates in cell movement and axonal guidance during embryonic development, as well as potentially regulating angiogenesis, inflammation, tissue remodeling, and cancer. Through interactions with its receptors, Netrin-1 plays varied roles in different physiological pathways, exerting different effects on various physiological and pathophysiological processes.

Summary: This study identifies Netrin-1 as an inflammation-inducible factor in the liver and reveals its contribution to hepatic inflammation through an atypical mechanism. Additionally, the study finds that an anti-Netrin-1 antibody has anti-inflammatory activity in hepatic inflammation.

Netrin-1 attenuates hepatic steatosis via UNC5b/PPAR?-mediated suppression of inflammation and ER stress

Sung Woo Choi et al.

Summary: This study investigated the role of netrin-1 in attenuating hepatic steatosis by suppressing inflammation and endoplasmic reticulum (ER) stress through PPAR gamma/auto-phagy-dependent signaling. The study found that netrin-1 can promote autophagy and PPAR gamma expression, leading to a reduction in inflammation and ER stress, lipid accumulation, and lipogenic proteins in hepatocytes. Furthermore, administration of netrin-1 reversed the effects of a high-fat diet on hepatic ER stress and lipid deposition. These findings shed light on the pathogenesis of hepatic steatosis in obesity and suggest a potential therapeutic approach for metabolic-associated fatty liver disease (MAFLD).

Disorganization and degeneration of liver sympathetic innervations in nonalcoholic fatty liver disease revealed by 3D imaging

Csaba Adori et al.

Summary: Research found that the hepatic nerves in NAFLD pathology show signs of degeneration, with chronic sympathetic hyperexcitation being a key factor in axonal degeneration. In experimental steatohepatitis, severe portal vein contraction was observed, spatially correlated with the extension of remaining nerves around the portal vein, providing insight into a potential intrahepatic neuronal mechanism of portal hypertension.
