Advancing the global public health agenda for NAFLD: a consensus statement

Upregulation of Nrf2 signaling and suppression of ferroptosis and NF-κB pathway by leonurine attenuate iron overload-induced hepatotoxicity

Summary: This study investigated the protective effects of the natural alkaloid leonurine against iron-induced hepatotoxicity in rats. The results showed that leonurine suppressed ferroptosis, oxidative damage, and inflammation in the liver tissues. It also enhanced the antioxidant potential and improved histopathological abnormalities. Mechanistically, leonurine modulated the Nrf2 and NF-kappa B signaling pathways. These findings highlight the hepatoprotective activity of leonurine against iron-induced hepatotoxicity.

Counteraction of Myocardial Ferritin Heavy Chain Deficiency by Heme Oxygenase-1

Summary: Given the important role of heme proteins in mitochondrial function, this study investigated the significance of myocardial ferritin heavy chain (FtH) in acute myocardial infarction. The results showed that FtH deletion did not impact mitochondrial regulation, bioenergetics, or cardiac function. Furthermore, FtH deletion led to the upregulation of anti-ferroptotic genes, including HO-1 and Slc7a11, suggesting a compensatory mechanism. The induction of HO-1 resulted in increased expression of Slc7a11, ultimately protecting the heart against ischemia-reperfusion-mediated ferroptosis and preserving myocardial and mitochondrial function.

Summary: The study found that ECH1 expression was significantly increased in human NASH biopsy specimens and in methionine choline-deficient (MCD) diet-fed mice. Overexpression of ECH1 alleviated hepatic steatosis, inflammation, fibrogenesis, apoptosis, and oxidative stress in mice, while also reducing alanine aminotransferase and proinflammatory cytokine levels in serum and triglyceride levels in livers. ECH1 may act as a potential therapeutic target for NASH by inhibiting ferroptosis.
