Targeting the chromatin remodeling protein BRG1 in liver fibrosis: Mechanism and translational potential

Targetable Brg1-CXCL14 axis contributes to alcoholic liver injury by driving neutrophil trafficking

Summary: In this study, we found that the Brg1 gene plays a crucial role in the development of alcoholic liver disease (ALD) and has potential implications for ALD intervention. Brg1 expression was increased in ALD mouse models, alcohol-exposed hepatocytes, and human ALD specimens. Manipulation of Brg1 expression in hepatocytes affected ALD development in mice. The deficiency of Brg1 specifically attenuated hepatic infiltration of Ly6G(+) neutrophils in ALD mice. Furthermore, the study identified CXCL14 as a potential target for Brg1, and manipulation of CXCL14 expression affected ALD pathogenesis.

Zinc finger transcription factor Egf1 promotes non-alcoholic fatty liver disease

Summary: Non-alcoholic fatty liver disease (NAFLD) is a contributing factor to the global epidemic of metabolic syndrome and is considered a prelude to end-stage liver diseases. In this study, the involvement of early growth response 1 (Egr1) in NAFLD was investigated, and it was found that Egr1 was upregulated in NAFLD. Further analysis revealed that serum response factor (SRF) mediated Egr1 transactivation. Depletion of Egr1 significantly mitigated NAFLD in mice. These findings identify Egr1 as a novel modulator of NAFLD and a potential target for intervention.

Eotaxin-1 expression is elevated in proliferating murine livers and can be stimulated by hepatocyte growth factor (HGF). Brg1 is a chromatin remodeling protein that directly binds to the eotaxin promoter to activate its transcription, and interacts with NF-kappa B/RelA to activate eotaxin transcription. Depletion of eotaxin or Brg1 deficiency delays liver regeneration.
