Alox15/15-HpETE Aggravates Myocardial Ischemia-Reperfusion Injury by Promoting Cardiomyocyte Ferroptosis

Summary: Myocardial ischemia-reperfusion (I/R) injury leads to cardiac dysfunction and cell loss. The time point and mechanisms of different types of cell death after reperfusion injury are unclear. This study found that ferroptosis is the predominant form of cell death during prolonged reperfusion and identified Alox15/15-HpETE as important regulators of cardiomyocyte ferroptosis.

Therapeutic potentials of cell death inhibitors in rats with cardiac ischaemia/reperfusion injury

Summary: Growing evidence suggests that ferroptosis, apoptosis, and necroptosis contribute to cardiac ischemia/reperfusion injury. This study found that apoptosis and ferroptosis, but not necroptosis, contribute to myocardial damage in acute cardiac ischemia/reperfusion injury. Inhibitors of these pathways provided effective cardioprotection through modulation of mitochondrial function and attenuation of apoptosis and ferroptosis.

The method allows for molecular and morphological profiling of cardiomyocytes from individual hearts of any postnatal age.

Inhibition of ELAVL1-mediated autophagic ferroptosis could be a potential new treatment strategy for myocardial I/R injury.

Elucidating the contribution of mitochondrial glutathione to ferroptosis in cardiomyocytes

Summary: Ferroptosis is a programmed iron-dependent cell death associated with lipid peroxidation, particularly phospholipids. Mitochondria play a crucial role in ferroptosis by being sensitive to ferroptotic stimuli and contributing to lipid peroxidation. The transport of reduced glutathione to mitochondria through DIC and OGC carrier proteins plays a key role in ferroptosis, and inhibiting these carriers can aggravate the process. Additionally, dihydrolipoic acid acts as an essential cofactor for mitochondrial enzymes and can attenuate ferroptosis by directly reducing peroxidized phospholipids.
