Rapamycin, Acarbose and 17α-estradiol share common mechanisms regulating the MAPK pathways involved in intracellular signaling and inflammation

Lily Wink et al.

Summary: Three drugs, ACA, Rapa, and 17aE2, have shown to regulate the ERK1/2 and p38-MAPK pathways. Effects of ERK1/2 signaling changes can be observed in both sexes, while the effects of 17aE2 are specific to males and consistent with lifespan effects. However, all three drugs inhibit age-related p38MAPK pathways and acute phase responses in both sexes, suggesting new approaches for preventing or reversing age-related inflammatory changes independent of lifespan effects.

Neuroprotective effects of Canagliflozin: Lessons from aged genetically diverse UM-HET3 mice

Hashan S. M. Jayarathne et al.

Summary: The aging brain is characterized by increased neuroinflammation and central insulin resistance, which contribute to neurodegenerative diseases and cognitive impairment. The anti-diabetes drug Canagliflozin (Cana) has shown neuroprotective effects in aged male mice by improving central insulin sensitivity, reducing gliosis in the hypothalamus, and improving exploratory and locomotor activity.

Molecular inhibition of RAS signalling to target ageing and age-related health

Mihails Laskovs et al.

Summary: The RAS/MAPK pathway plays a crucial role in cancer and is also associated with ageing. Decreasing RAS/MAPK signaling activity can extend lifespan, improve healthspan, and delay or prevent age-related functional decline. Evidence suggests that repurposing small molecule inhibitors of RAS/MAPK signaling can enhance health during ageing, but challenges need to be addressed before clinical implementation.

Insights Into the Links Between Proteostasis and Aging From C. elegans

William Hongyu Zhang et al.

Summary: Proteostasis is crucial for organism longevity and health, with Caenorhabditis elegans serving as a valuable model organism for studying proteostasis in the context of aging, longevity, and disease. The insights gained from C. elegans research offer potential therapeutic targets for delaying aging and preventing age-related diseases.

Cap-independent translation: A shared mechanism for lifespan extension by rapamycin, acarbose, and 17α-estradiol

Ziqian Shen et al.

Summary: The study suggests that rapamycin and acarbose can prevent age-related declines in cap-independent translation (CIT) target proteins in both sexes, while 17 alpha-estradiol has a similar effect only in males. The activity of mTORC1 is blocked by rapamycin, acarbose, and 17 alpha-estradiol in age-related increases. The increase in METTL3 activity and CIT-dependent proteins may represent a shared pathway for both long-lived mutant mice and drug-induced lifespan extension in mice.
