DAVID: a web server for functional enrichment analysis and functional annotation of gene lists (2021 update)

Brad T. Sherman et al.

Summary: DAVID, a popular bioinformatics resource system, has undergone updates in 2021, including the rebuilding of the DAVID Gene system, increasing the number of gene-term records for annotation types, adding new annotations, assigning specific subgroups, and adding a species parameter for improved user experience.

Molecular annotation of extramedullary acute myeloid leukemia identifies high prevalence of targetable mutations

Somedeb Ball et al.

Summary: The study revealed frequent targetable mutations in EM-AML patients, with EM-site NGS being recommended for selecting potential targeted therapies for these patients. Targetable alterations such as IDH1, NPM1, IDH2, FLT3, and KMT2A-PTD were identified in a significant proportion of patients, and treatment with IDH inhibitors based on EM-site NGS showed promising results in achieving complete responses.

The cytokine network in acute myeloid leukemia

Michela Luciano et al.

Summary: Acute myeloid leukemia (AML) is a heterogeneous blood cancer with complex disease characteristics and cytokine networks, making treatment challenging. The development of new targeted therapies and understanding the functions of cytokines are important for improving treatment options.

Genomic analysis in myeloid sarcoma and comparison with paired acute myeloid leukemia

Nancy Y. Greenland et al.

Summary: Myeloid sarcoma is a rare manifestation of acute myeloid leukemia characterized by extramedullary proliferation of myeloid blasts. Studies have shown that the genetic profiles observed in myeloid sarcoma are generally similar to paired acute myeloid leukemia samples, suggesting a common precursor origin for these tumors rather than de novo tumors. Additionally, myeloid sarcoma cases with a higher number of single-nucleotide variants tend to have worse clinical outcomes. The identification of these abnormalities may contribute to improved prognostic classification and the discovery of new therapeutic targets for myeloid sarcoma.

Myeloid sarcoma, chloroma, or extramedullary acute myeloid leukemia tumor: A tale of misnomers, controversy and the unresolved

Rory M. Shallis et al.

Summary: The classification and definition of myeloid sarcoma by the World Health Organization is imprecise, with a more accurate term being extramedullary acute myeloid leukemia tumor (eAML). eAML is associated with aberration of cellular adhesion molecules, chemokine receptors/ligands, and RAS-MAPK/ERK signaling. Diagnosis requires tissue biopsy, and treatment may involve both local therapy and systemic anti-leukemia therapy.

Newly diagnosed isolated myeloid sarcoma-paired NGS panel analysis of extramedullary tumor and bone marrow

Nils W. Engel et al.

Summary: Isolated myeloid sarcoma is a rare malignancy where tumors form in various locations while the bone marrow remains unaffected. Clonal evolution and clonal hematopoiesis of indeterminate potential were observed in some cases with detectable mutations. Clinical presentation of patients suggested possible extramedullary blast trafficking to distal sites, avoiding the bone marrow.
