Apoptosis reprogramming triggered by splicing inhibitors sensitizes multiple myeloma cells to Venetoclax treatment

Debora Soncini et al.

Summary: Recent studies have found aberrant splicing in cancer cells and suggested it as a potential therapeutic strategy. This study evaluated the expression of spliceosome machinery components in multiple myeloma cells and found that modulating splicing can impair cell growth and survival. The findings suggest that targeting splicing could make multiple myeloma patients more vulnerable to BCL2 inhibitors.

Targeting the spliceosome machinery: A new therapeutic axis in cancer?

Beatrice Eymin

Summary: Pre-mRNA splicing plays a critical role in regulating gene expression in eukaryotic cells, with over 90% of protein-encoding transcripts being alternatively spliced, increasing protein diversity. Abnormal splice variants identified in cancers have led to a new anti-cancer strategy targeting the spliceosome machinery, though the potential and efficacy of these therapies are still under debate.

Monitoring of clonal evolution of acute myeloid leukemia identifies the leukemia subtype, clinical outcome and potential new drug targets for post-remission strategies or relapse

Esther Onecha et al.

Summary: In cases of treatment failure in acute myeloid leukemia (AML), mutational profiling can help differentiate primary refractoriness from relapse, providing insights for treatment guidance and prognostic risk assessment. Additionally, molecular follow-up using a custom NGS myeloid panel can aid in monitoring treatment response, identifying minimal residual disease markers, guiding post-remission strategies, and selecting new drugs for leukemia relapse.

Global Proteomic Profiling of Pediatric AML: A Pilot Study

Nam H. K. Nguyen et al.

Summary: Acute myeloid leukemia (AML) is a heterogeneous disease with poor prognosis, and the current study focused on investigating proteomic differences between subtypes of AML, providing insights into the physiology of AML subtypes and potential druggable proteome targets. Most biological functions within cells are operated by proteins, highlighting the significance of studying proteomics in drug development.
