Macrophages at the interface of the co-evolving cancer ecosystem

Summary: Macrophages play versatile and heterogeneous roles in innate immunity, maintaining immune responses and tissue repair. However, when co-opted by malignant outgrowth, macrophages can contribute to tumor evolution within the tumor microenvironment. This review explores the sources of influence that shape tumor-associated macrophage (TAM) phenotypic states in solid neoplasms and discusses how TAMs steer tumor evolution in response to these signals. The authors propose new therapeutic approaches to exploit the complex roles of TAMs in cancer.

PD-L1 checkpoint blockade promotes regulatory T cell activity that underlies therapy resistance

Summary: Despite the success of immune checkpoint blockade (ICB) in some cancer types, most patients do not respond well and develop resistance to ICB. This study identified enhanced suppressive capacity of regulatory T cells (T-reg cells) in therapy-resistant tumors. T-reg cell depletion reversed resistance and increased effector T cells. Similar findings were observed in human patients, suggesting that targeting T-reg cells is important to enhance therapeutic efficacy.

Tumour-infiltrating lymphocytes in oropharyngeal cancer: a validation study according to the criteria of the International Immuno-Oncology Biomarker Working Group

Summary: This study evaluated the prognostic significance of tumor-infiltrating lymphocytes (TILs) in oropharyngeal squamous cell carcinoma (OPSCC). The results showed that TILs were significantly associated with disease-specific survival and overall survival, and had prognostic value in both HPV-positive and HPV-negative cases.

Impact of human papillomavirus on the tumor microenvironment in oropharyngeal squamous cell carcinoma

Summary: This study identified differences in tumor microenvironment (TME) associated with HPV infection in OPSCC, with enrichment of specific cell types in HPV+ and HPV- OPSCCs. By constructing a coexpression network, it was found that HPV+ OPSCCs exhibit elevated immune activities and suppressed extracellular matrix-related activities compared to HPV- OPSCCs. Clinical analysis showed that TME-relevant genes were closely linked to prognosis and therapy response in OPSCC, with results validated in an independent OPSCC cohort.

Hallmarks of Cancer: New Dimensions

Summary: The concept of the hallmarks of cancer is a tool for simplifying the complexity of cancer phenotypes and genotypes into basic principles. This article proposes phenotypic plasticity and disrupted differentiation as distinct hallmark capabilities, and discusses nonmutational epigenetic reprogramming and polymorphic microbiomes as enabling characteristics. Additionally, the importance of senescent cells in the tumor microenvironment is highlighted. The integrative concept of the hallmarks of cancer helps in understanding the mechanisms of cancer development and applying that knowledge to cancer medicine.

Summary: The expression of CD4+CD161+ effector memory T cells is associated with improved survival in HPV16+ oropharyngeal squamous cell carcinoma. Therapeutic vaccination activates and expands CD4+CD161+ effector T cells. CD161 functions dynamically and is regulated by cell intrinsic and extrinsic factors. CD161 expressing CD4+ T cells respond more vigorously to suboptimal antigen stimulation and may amplify TCR signals.

Future clinical trials considering LAG-3, TIM-3, and VISTA expression are necessary to further explore the efficacy of checkpoint blockade therapy.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
