Local Bilayer Hydrophobicity Modulates Membrane Protein Stability

Summary: This study determined an empirical correlation between the surface area of nonpolar side chains, transfer free energies, and local water concentration in the membrane, allowing for accurate estimation of Delta G(sc)(0) at any location in the bilayer. Additionally, the calculated interface-to-bilayer transfer free energy values were found to be similar to biological translocon-based transfer free energies, suggesting that the translocon mimics the bilayer interface energetically.

Lipids modulate the BH3-independent membrane targeting and activation of BAX and Bcl-xL

Summary: The regulation of apoptosis involves the targeting of Bcl-2 proteins to the mitochondrial outer membrane, with BAX and Bcl-xL found to interact with membranes in the presence of cellular concentrations of Mg2+/Ca2+ even in cells lacking BH3 effectors. This targeting and activation mechanism provides a mechanistic explanation for the involvement of Bcl-2 proteins in mitochondrial processes, highlighting the potential amplification of apoptotic signaling and counteraction against premature commitment to apoptosis.

Membrane-selective nanoscale pores in liposomes by a synthetically evolved peptide: implications for triggered release

Summary: Macrolittins are a novel family of alpha-helical pore-forming peptides that show unprecedented selectivity for releasing macromolecular cargoes from phosphatidylcholine (PC) liposomes over cell plasma membranes. This selectivity is partly explained by the sensitivity of macrolittin activity to physical chemical properties of the bilayer hydrocarbon core. In the presence of cells, macrolittins release vesicle-entrapped cargoes without directly affecting the cells or involving vesicle-vesicle or vesicle-cell interactions.
