KLF5/MDM2 Axis Modulates Oxidative Stress and Epithelial-Mesenchymal Transition in Human Lens Epithelial Cells: The Role in Diabetic Cataract

PM2.5 increases susceptibility to acute exacerbation of COPD via NOX4/Nrf2 redox imbalance-mediated mitophagy

Summary: The increasing abundance of PM2.5 in the environment has increased susceptibility to AECOPD. This study evaluated the role of NOX4/Nrf2 redox imbalance on AECOPD induced by PM2.5-CS exposure and found that PM2.5 exacerbates cytotoxicity by enhancing NOX4/Nrf2 redox imbalance-mediated mitophagy. Restoring the NOX4/Nrf2 redox balance can alleviate this cytotoxicity and ameliorate AECOPD.

TGF-β/Smad signaling pathway plays a crucial role in patulin-induced pro-fibrotic changes in rat kidney via modulation of slug and snail expression

Summary: In this study, the mechanism involved in pro-fibrotic changes in the kidney after low-dose chronic exposure to PAT was investigated using in vitro and in vivo approaches. The results demonstrated that PAT exposure caused the generation of reactive oxygen species (ROS), activation of MAPKs and cJun/Fos signaling pathways, and induction of TGF-β(1) expression. Additionally, it was found that PAT-induced TGF-β(1) further activated the TGF-β(1)/smad signaling pathways and modulated the expression of slug and snail, leading to the regulation of pro-fibrotic molecules. The in vivo results also confirmed the kidney injury/toxicity effects of PAT exposure in rats.

KLF5 Is Induced by FOXO1 and Causes Oxidative Stress and Diabetic Cardiomyopathy

Summary: The study revealed that diabetic cardiomyopathy (DbCM) is associated with increased cardiac expression of KLF5, which is regulated by FOXO1. KLF5 induces DbCM by promoting oxidative stress through NOX4 and ceramide accumulation in the heart. Inhibiting KLF5 could alleviate superoxide formation and improve cardiac function in diabetic mice.

Mdm2 phosphorylation by Akt regulates the p53 response to oxidative stress to promote cell proliferation and tumorigenesis

Summary: The phosphorylation of Mdm2 by Akt was found to reduce p53 levels and prevent senescence in cells exposed to oxidative stress. This resulted in the promotion of lung cancers driven by Kras(G12D) and carcinogen-induced papilloma and hepatocellular carcinomas in mouse models of ROS-induced cancer. These findings highlight a unique physiological role for Akt-Mdm2-p53 signaling in regulating cell growth and tumorigenesis in response to oxidative stress.

KLF5-mediated Eppk1 expression promotes cell proliferation in cervical cancer via the p38 signaling pathway
