Development of an HiBiT-tagged reporter H3N2 influenza A virus and its utility as an antiviral screening platform

Chengcheng Zhang et al.

Summary: In this study, a reporter influenza A/H3N2 virus (A/NY-HiBiT) derived from a clinical isolate was constructed by placing a minimized HiBiT tag to the viral nuclear-export protein. The HiBiT tag did not affect the viral genome balance, and the recombinant A/NY-HiBiT virus remained stable. The replication profile of the HiBiT-tagged virus could be measured using a simple Nano-Glo assay, providing a robust platform for high-throughput screening. Using this platform, three fractions from Chinese medicinal materials were identified as potent anti-influenza A virus actives.

Soluble ORF2 protein enhances HEV replication and induces long-lasting antibody response and protective immunity in vivo

Philipp Ralfs et al.

Summary: This study investigates the role of ORF2s in HEV replication and antibody generation. The findings suggest that ORF2s may not be essential for viral replication in vivo but is required for the formation of long-lived antibody responses and protection against HEV re-exposure.

Summary: HCV-1, a major cause of acute viral hepatitis, has been difficult to study due to its poor replication in cultured cells. A cDNA clone was successfully constructed and an infectious virus was produced, providing a useful tool for studying HCV-1 replication and severe hepatitis in infected pregnant women, as well as for developing safer treatment options.

Expanding the Hepatitis E Virus Toolbox: Selectable Replicons and Recombinant Reporter Genomes

Noemie Oechslin et al.

Summary: Hepatitis E virus (HEV) is now considered as one of the most frequent causes of acute hepatitis worldwide, but it has received relatively little attention for decades. Recent advances in the molecular virology of HEV have allowed study of the entire viral life cycle and exploration of host factors required for infection.

Summary: Bioluminescent reporter viruses are important tools in molecular virological research and have been widely used in studying viral life cycles and developing antiviral drugs. This study developed the HEV-nanoKAZ system and successfully applied it in drug screening to identify four hit drugs with anti-HEV activity.

The Capsid (ORF2) Protein of Hepatitis E Virus in Feces Is C-Terminally Truncated

Takashi Nishiyama et al.

Summary: The study compared the molecular size of the ORF2 protein among fecal HEV, cell-culture-generated HEV, and treated HEVcc, revealing that the ORF2 proteins associated with fecal HEV were C-terminally truncated and showed the same size as treated HEVcc. This sheds light on the maturation of nonenveloped HEV and is important for understanding the HEV lifecycle.

Summary: This study aims to elucidate the replication step of Hepatitis E virus (HEV) and identify the replication location within host cells. By inserting epitope tags and conducting protein analysis, we discovered the significant role of ORF1 polyprotein in the cell nucleus and perinuclear substructures and identified potential replication factories for HEV.

Ritonavir Blocks Hepatitis E Virus Internalization and Clears Hepatitis E Virus In Vitro with Ribavirin

Putu Prathiwi Primadharsini et al.

Summary: Combination therapy with ritonavir and ribavirin significantly improves the inhibition of hepatitis E virus (HEV) growth, even in cells with robust virus production. This study provides insight into the potential of using drugs that block different steps in the HEV life cycle as a novel treatment strategy for chronic hepatitis E.

Treatment for chronic hepatitis E virus infection: A systematic review and meta-analysis

Myrte Gorris et al.

Summary: According to a systematic review, treatment with ribavirin can achieve a sustained virological response (SVR) in 78% of patients, with side effects such as anemia requiring dose reduction or blood transfusion in 37% of patients. For chronic hepatitis E, ribavirin should be considered as the first treatment step. Treatment with pegylated interferon-alpha increases the risk of transplant rejection and should be administered with caution.

Structural and molecular biology of hepatitis E virus

Bo Wang et al.

Summary: Hepatitis E virus (HEV) is an emerging zoonotic pathogen with a broad host range, mainly transmitted by fecal-oral route. The genome of HEV typically contains three open reading frames encoding different proteins. Research on the structural and molecular biology of HEV, including virion structure, genomic organization, RNA structures, viral proteins and their functions, and the life cycle of HEV, is ongoing with gaps in understanding still remaining.
