DHA and EPA inhibit porcine coronavirus replication by alleviating ER stress

Porcine epidemic diarrhea virus activates PERK-ROS axis to benefit its replication in Vero E6 cells

Summary: This study demonstrates that porcine epidemic diarrhea virus (PEDV) infection triggers unfolded protein response (UPR) in Vero E6 cells by activating the PERK/eIF2 alpha pathway, leading to increased expression of proapoptotic protein CHOP and ER oxidoreductase 1 alpha. Inhibition of PERK or CHOP reduces expression of ERO1 alpha and generation of ROS, while inhibition of ERO1 alpha decreases PEDV-induced ROS generation. Genetic or pharmacological inhibition of PERK, CHOP, ERO1 alpha, or ROS suppresses PEDV replication. These findings provide evidence that PEDV manipulates endoplasmic reticulum signaling to disrupt redox homeostasis and promote its replication.
