CRISPR/Cas mediated epigenome editing for cancer therapy

Structural basis for mismatch surveillance by CRISPR-Cas9

Summary: In this study, the structure of Cas9 during mismatch cleavage was determined using kinetics-guided cryo-electron microscopy. It was found that a linear conformation of the guide RNA-DNA duplex formed in the presence of mismatches, preventing Cas9 activation. Additionally, mismatches distal to the protospacer adjacent motif were stabilized by reorganization of a loop in the RuvC domain. Mutations of mismatch-stabilizing residues reduced off-target DNA cleavage while maintaining rapid on-target DNA cleavage. This study provides proof of concept for the design of next-generation high-fidelity Cas9 variants targeting mismatch tolerance regions.

A humanized knockin mouse model of Duchenne muscular dystrophy and its correction by CRISPR-Cas9 therapeutic gene editing

The model successfully restored dystrophin expression and improved pathological features and muscle strength. This model provides a valuable tool for assessing clinically relevant gene editing strategies and advancing the therapeutic translation of CRISPR-Cas9 gene editing for DMD correction.

Targeting histone acetylation dynamics and oncogenic transcription by catalytic P300/CBP inhibition

Summary: The study found that catalytic inhibition of P300/CBP dynamically disrupts steady-state acetylation kinetics and suppresses oncogenic transcriptional networks without changing chromatin accessibility. CRISPR-Cas9 screening identified NCOR1 and HDAC3 as the key antagonists of P300/CBP by counteracting acetylation turnover kinetics. Deacetylation of H3K27 provides nucleation sites for reciprocal methylation switching, which can be therapeutically exploited by simultaneous KDM6A and P300/CBP inhibition.
