Remembering your A, B, C's: Alzheimer's disease and ABCA1

Cutler T. Lewandowski et al.

Summary: ATP binding cassette protein A1 (ABCA1) plays a central role in cholesterol mobilization. Reduced ABCA1 expression or activity is associated with Alzheimer's disease and other disorders. Current therapeutic approaches to enhance ABCA1 activity have not been successfully translated into clinical practice. The risk factors for Alzheimer's disease development and progression, such as type 2 diabetes and cardiovascular disease, highlight the intersection of cholesterol transport and inflammation. Upregulation of ABCA1 can have positive effects on APOE lipidation, insulin sensitivity, peripheral vascular and blood-brain barrier integrity, and anti-inflammatory signaling.

Neuropathological consensus criteria for the evaluation of Lewy pathology in post-mortem brains: a multi-centre study

Johannes Attems et al.

Summary: Several staging systems exist for the neuropathological diagnosis of LBD, with varying levels of inter-rater reliability. The LPC system, based on a dichotomous approach for scoring LP, showed good reproducibility and allowed for the classification of all cases into distinct categories. It may serve as a reliable and useful standard future approach for the post-mortem evaluation of LP.

NOTCH3 variants are more common than expected in the general population and associated with stroke and vascular dementia: an analysis of 200 000 participants

Bernard P. H. Cho et al.

Summary: Cysteine-altering NOTCH3 variants, previously associated with CADASIL, are more common in the general population than expected, and are risk factors for stroke and vascular dementia. These variants are also associated with MRI changes indicative of small vessel disease.

Summary: This study identified multiple pathogenic variants of NOTCH3, confirming its pathological role in SVaD, but found no association with AD.
