Potential cerebrovascular accident signal for risankizumab: A disproportionality analysis of the FDA Adverse Event Reporting System (FAERS)

Richard H. Woods

Summary: This study found a previously unreported signal of disproportionate cerebrovascular accident (CVA) reporting associated with the real-world use of risankizumab for the treatment of plaque psoriasis. Further long-term observational data is necessary to better characterize this unconfirmed potential safety signal.

Interleukin-17 Links Inflammatory Cross-Talks Between Comorbid Psoriasis and Atherosclerosis

Yan Wang et al.

Summary: Psoriasis is associated with an increased risk of cardiovascular disease. Clinical studies have shown that IL-17A inhibitors can effectively improve both psoriasis and atherosclerosis symptoms, suggesting that IL-17A may play a crucial role in the comorbidity of these two diseases.

Apremilast exerts protective effects on stroke outcomes and blood-brain barrier (BBB) dysfunction through regulating Rho-associated protein kinase 2 expression

Mingyuan Wang et al.

Summary: In this study, the researchers investigated the effects of APR on stroke outcomes and blood-brain barrier (BBB) dysfunction using a mouse model of middle cerebral artery occlusion (MCAO). The results showed that APR attenuated neurological injury, reduced infarct size, and improved grip strength in MCAO mice. APR treatment also helped restore BBB function and reduce neuroinflammation. In vitro experiments demonstrated that APR improved endothelial cell permeability and restored the expression of Claudin-5. These findings suggest that APR has potential as a therapeutic intervention for ischemic stroke by targeting BBB dysfunction and neuroinflammation.
