Targeting cellular senescence with senotherapeutics: senolytics and senomorphics

Summary: Chronologic age is the main risk factor for coronary artery disease, and age-related subclinical leukemogenic mutations in blood cells have been found to play a surprising role in this disease. These mutations occur in genes such as DNMT3A, TET2, ASXL1, and JAK2. Studies on mice and humans have shown the involvement of inflammatory pathways in linking clonal hematopoiesis with coronary artery disease. Further analysis using larger, longitudinal, and multiomics approaches is providing mechanistic insights. These observations have expanded our understanding of the genetic basis of coronary artery disease, connecting it with hematologic neoplasia. They also have implications for the development of novel precision medicine approaches for this disease.

Hallmarks of aging: An expanding universe

These hallmarks are interconnected with each other and with the recently proposed hallmarks of health.

Effect of long-term caloric restriction on DNA methylation measures of biological aging in healthy adults from the CALERIE trial

Summary: The geroscience hypothesis suggests that slowing down or reversing molecular changes associated with aging can delay or prevent chronic diseases and extend healthy lifespan. A post hoc analysis of the CALERIE trial showed that caloric restriction intervention can slow down the pace of aging, but did not significantly affect biological age estimates. Despite small effect sizes, even modest slowdown of aging can have profound effects on population health. This finding supports the geroscience hypothesis and provides a basis for future long-term follow-up trials to establish the effects of intervention on primary healthy-aging endpoints.

Summary: This study finds that hematopoietic mosaic loss of Y chromosome (mLOY) is associated with increased risk of mortality and age-related diseases in men. The animal experiment results show that male mice reconstituted with bone marrow cells lacking the Y chromosome display increased mortality and age-related profibrotic pathologies including reduced cardiac function. Further research reveals that cardiac macrophages lacking the Y chromosome exhibit polarization toward a more fibrotic phenotype, and treatment with a specific antibody can alleviate cardiac dysfunction. A prospective study also reveals that mLOY in blood is associated with an increased risk for cardiovascular disease and heart failure-associated mortality.

Lifespan benefits for the combination of rapamycin plus acarbose and for captopril in genetically heterogeneous mice

Summary: In this study, different drugs were tested on mice for their effects on lifespan. The combination of rapamycin and acarbose showed a longer lifespan in male mice compared to the previous cohorts, indicating its stronger potency. Captopril led to a small increase in female lifespan. Other drugs tested did not show any significant lifespan benefits. Combinations of anti-aging drugs may have superior effects than using a single drug alone. Further studies on captopril at different dosages are recommended.
