Modulating Angiogenesis by Proteomimetics of Vascular Endothelial Growth Factor

Summary: New sulfono-gamma-AA peptide-based helical peptidomimetics were designed to mimic a key binding interface on VEGF for VEGFR recognition. These peptides, specifically V2 and V3, exhibited different effects on angiogenesis in vitro, with V3 potently inhibiting angiogenesis and V2 activating angiogenesis, possibly due to their selective binding to VEGFR-1 and VEGFR-2, respectively.

The folding propensity of α/sulfono-γ-AA peptidic foldamers with both left- and right-handedness

Summary: This study demonstrated the solid-state right-handed helical propensity of the 4(13)-helix of L-alpha/L-sulfono-gamma-AA peptides, even as short as 11-mer, using high-resolution X-ray single crystallography. The atomic level folding conformation of the foldamers was elucidated by 2D NMR and circular dichroism, showing that the helical handedness of this class of foldamer is controlled by the chirality of their chiral side chains.

Discovery of Cyclic Peptidomimetic Ligands Targeting the Extracellular Domain of EGFR

Summary: A novel small molecule, M-2-5, was discovered to tightly bind to the extracellular domain of EGFR and effectively antagonize EGF-stimulated EGFR phosphorylation and downstream signal transduction. Furthermore, it showed remarkable resistance to proteolytic degradation, effectively inhibiting cell proliferation and migration in vitro and suppressing tumor growth in vivo.
