Enzyme-Instructed Peptide Assembly Favored by Preorganization for Cancer Cell Membrane Engineering

Summary: An enzyme-instructed peptide self-assembly (EISA) strategy is used to selectively modify cancer cell membranes, and different phosphorylation patterns in phosphopeptides targeting EGFR can impact their self-assembly abilities and binding affinity for EGFR. The phosphorylated peptide pY1 shows the highest capability for preorganization and the fastest dephosphorylation in the presence of alkaline phosphatase (ALP), making it the most effective at constructing stable peptide assemblies on cancer cell membranes. Additionally, peptide-protein and peptide-peptide co-assembly strategies are employed to apply antigens to cancer cell membranes, offering a potential technique for cancer immunotherapy.

Antiviral supramolecular polymeric hydrogels by self-assembly of tenofovir-bearing peptide amphiphiles

Summary: The development of long-acting antiviral therapeutic delivery systems is crucial for improving the treatment and prevention of HIV and chronic HBV. This study reports the conjugation of tenofovir to rationally designed peptide amphiphiles to create antiviral prodrug hydrogelators. The resulting conjugates can self-assemble into one-dimensional nanostructures and undergo rapid gelation to form a drug depot. Our findings suggest that minor changes in peptide design can affect gelation and release.

Supramolecular Filament Hydrogel as a Universal Immunomodulator Carrier for Immunotherapy Combinations

Summary: A peptide-based supramolecular filament hydrogel was designed and synthesized as a carrier for localized delivery of three immunomodulating agents. The hydrogel acted as a depot for sustained release of the immunotherapeutic agents, resulting in enhanced antitumor activities and reduced side effects. Combination therapy using the hydrogel led to complete regression of tumors and elicited a systemic antitumor immunity. This hydrogel offers a simple and generalizable strategy for local delivery of immunomodulators, improving treatment outcomes.

Self-assembling paclitaxel-mediated stimulation of tumor-associated macrophages for postoperative treatment of glioblastoma

Summary: We report on a self-assembling paclitaxel filament hydrogel that stimulates macrophage-mediated immune response for local treatment of recurrent glioblastoma. The paclitaxel filaments elicit an immune-stimulating tumor microenvironment and sensitizes tumor to the aCD47-mediated blockade, promoting tumor cell phagocytosis by macrophages and triggering an antitumor T cell response. As adjuvant therapy after surgery, this aCD47/PF supramolecular hydrogel effectively suppresses primary brain tumor recurrence and prolongs overall survivals.
