Success probability of high-affinity DNA aptamer generation by genetic alphabet expansion

Summary: Nucleic acid aptamers, as antibody alternatives, can specifically bind to target molecules. These aptamers are generated by isolating candidates from libraries with random sequence fragments, through an evolutionary engineering system. The affinity of DNA aptamers to target proteins can be increased over 100-fold by incorporating hydrophobic unnatural bases (UBs) into the library.
