Copper-Catalyzed Electrophilic Amination: An Umpolung Strategy for New C -N Bond Formations

Koji Hirano

Summary: The development of new C-N bond forming strategies is crucial due to the importance of nitrogen atom in bioactive molecules and functional materials. This account presents the nitrogen-umpolung-enabled copper-catalyzed highly selective amination protocols developed by the author's research group. The application of these methods to the synthesis of alkyl-amines with rich functionality is demonstrated.

Cooperative Pd/Cu Catalysis for Alkene Arylboration: Opportunities for Divergent Reactivity

Stanna K. Dorn et al.

Summary: Controlling selectivity is a major challenge in alkene difunctionalization. This study highlights a Pd/Cu-cooperative catalytic system for alkene arylboration that enables controlled introduction of substituents. Divergent reactivity from a single substrate can be achieved by tuning catalysts and reaction conditions.

anti-Selective synthesis of β-boryl-α-amino acid derivatives by Cu-catalysed borylamination of α,β-unsaturated esters

Soshi Nishino et al.

Summary: A copper-catalyzed reaction has been developed for the regio- and diastereoselective borylamination of alpha,beta-unsaturated esters with B(2)pin(2) and hydroxylamines. The reaction delivers acyclic beta-boryl-alpha-amino acid derivatives with high anti-diastereoselectivity, which is difficult to achieve using established methods. Furthermore, the use of a chiral phosphoramidite ligand allows for the enantioselective synthesis of optically active beta-borylated alpha-amino acids. The resulting products can be further transformed into beta-functionalized alpha-amino acids, which are of great interest in medicinal chemistry.

Palladium-Catalyzed Remote Diborylative Cyclization of Dienes with Diborons via Chain Walking

Shota Kanno et al.

Summary: A novel catalytic method using chain walking and dimetal reagents was developed for the remote bismetalation of alkene substrates, resulting in cyclopentane derivatives with two boryl groups at remote positions. The regioselective transformation of an unactivated sp(3) C-H bond to a C-B bond successfully achieved the sequential construction of three distant bonds for 1,n-dienes (n >= 7).

α-Boryl Organometallic Reagents in Catalytic Asymmetric Synthesis

Chenlong Zhang et al.

Summary: In recent years, a-boryl organometallic reagents have become popular as versatile nucleophiles in asymmetric synthesis, enabling chemo- and stereoselective coupling reactions to produce enantioenriched boronic esters. This allows for efficient construction of complex structures with high functional group compatibility. Efforts have been focused on preparing enantiomerically enriched a-boryl organometallic reagents and developing stereoselective reactions of related racemic materials.
