New perspectives on the molecular mechanisms of stress signalling by the nucleotide guanosine tetraphosphate (ppGpp), an emerging regulator of photosynthesis in plants and algae

Marwa Mehrez et al.

Summary: This review discusses the role of GTP and ppGpp in prokaryotes and eukaryotes, focusing on their function as regulators of photosynthetic activity in plant and algal chloroplasts, as well as their emerging roles in environmental stress acclimation. The authors highlight the gaps in our understanding of the molecular mechanisms of GTP and ppGpp signaling in plants and algae, and propose that filling in these gaps may reveal conserved and plant- and algal-specific signaling mechanisms that contribute to chloroplast regulation and stress tolerance.

Metabolic changes contributing to large biomass production in the Arabidopsis ppGpp-accumulating mutant under nitrogen deficiency

Mina Goto et al.

Summary: The Arabidopsis ppGpp-overproducing mutant shows a larger biomass than wild type under nitrogen-limiting conditions by modulating amino-acid metabolism. The regulatory nucleotide ppGpp controls gene expression and enzyme activities, but the mechanisms behind its influence on plant growth and biomass are still unclear. The accumulation of ppGpp affects amino-acid and protein homeostasis, facilitating growth under nitrogen-limiting conditions.

Regulation of ppGpp Synthesis and Its Impact on Chloroplast Biogenesis during Early Leaf Development in Rice

Kazuhiro Ito et al.

Summary: Guanosine tetraphosphate (ppGpp) is involved in regulating chloroplast gene expression in plants, affecting photosynthesis and plant growth. In rice, ppGpp inhibits GTP biosynthesis enzyme GKpm, and overexpression of ppGpp synthase RSH3 enhances chlorosis and lowers photosynthetic rates. The basal ppGpp level in rice during leaf development appears to be determined by the opposing activities of multiple RSH enzymes.

Guanosine tetraphosphate (ppGpp) accumulation inhibits chloroplast gene expression and promotes super grana formation in the moss Physcomitrium (Physcomitrella) patens

Seddik Harchouni et al.

Summary: This study investigates the function of ppGpp signaling in the moss Physcomitrium patens and reveals that ppGpp accumulation inhibits chloroplast gene expression, resulting in a drop in photosynthetic capacity, but does not affect plant growth. This suggests that bryophytes and vascular plants may have different tolerances to defects in photosynthesis, potentially reflecting their different growth strategies.

Two P or Not Two P: Understanding Regulation by the Bacterial Second Messengers (p)ppGpp

Gert Bange et al.

Summary: Under stressful growth conditions and nutrient starvation, bacteria adapt by synthesizing signaling molecules that profoundly reprogram cellular physiology, with (p)ppGpp playing a key role in this process. The synthesis and hydrolysis of this signaling molecule are carried out by specific enzymes, directly impacting intracellular effectors. Studies have revealed both common features and differences in the effects of (p)ppGpp on gram-negative and gram-positive bacteria.

ITPK1 is an InsP6/ADP phosphotransferase that controls phosphate signaling in Arabidopsis

Esther Riemer et al.

Summary: This study provides comprehensive insights into how inositol (pyro)phosphate metabolism is regulated by Pi and dependent on the inositol phosphate kinase ITPK1. The research demonstrates that inositol pyrophosphates are more responsive to Pi than lower inositol phosphates and can be separated in different isomers in plants. Additionally, the study identifies new regulatory inositol pyrophosphates and their synthesis with Pi-dependent changes in nutritional and energetic states.
