Arsenic in medicine: past, present and future

Ngozi P. Paul et al.

Summary: Arsenicals, one of the oldest treatments for various human disorders, have been paradoxically used as therapeutic agents since ancient times despite their toxicity. After the discovery of antibiotics, most arsenic-based drugs were abandoned, but recent years have seen a renewed interest in their clinical use. The current applications of arsenicals include antimicrobial, antiviral, antiparasitic, and anticancer uses. With the rise of antibiotic resistance and emerging pathogens, revisiting arsenicals as an effective treatment option is suggested.

Characterization and Mechanistic Study of the Radical SAM Enzyme ArsS Involved in Arsenosugar Biosynthesis

Jinduo Cheng et al.

Summary: The study characterizes ArsS as a DDMAA synthase, revealing a key step in arsenosugar biosynthesis. Phylogenomic and biochemical analyses show that DDMAA synthases are widely distributed in different bacterial phyla, likely originating from cyanobacteria. This highlights the catalytic diversity of radical SAM enzymes.

clinker & clustermap.js: automatic generation of gene cluster comparison figures

Cameron L. M. Gilchrist et al.

Summary: Genes involved in biological pathways are often found in gene clusters, and comparing these clusters can provide valuable insights into their function and evolutionary history. To simplify the tedious process of comparing and visualizing gene cluster similarity, a Python tool called clinker and a companion JavaScript library called clustermap.js have been developed to automatically generate accurate, interactive, publication-quality gene cluster comparison figures from sequence files.

Transcriptional and Post-Transcriptional Polar Effects in Bacterial Gene Deletion Libraries

Andre Mateus et al.

Summary: Study shows that single-gene deletions in bacterial operons can have transcriptional and post-transcriptional effects on other genes, with nearly 40% of operons exhibiting more than 2-fold up- or downregulation. Additionally, a post-transcriptional effect leads to downregulation of the gene directly downstream of the deletion.

Identification of the Biosynthetic Gene Cluster for the Organoarsenical Antibiotic Arsinothricin

Adriana E. Galvan et al.

Summary: The soil bacterium Burkholderia gladioli GSRB05 produces the broad-spectrum antibiotic arsinothricin, with a biosynthetic gene cluster consisting of three genes identified. The biosynthesis and transport of arsinothricin only require three genes, in contrast to the over 20 genes needed for the phosphonate counterpart. This discovery may lead to the development of more effective organoarsenical antibiotics and shed light on the complexity of the arsenic biogeochemical cycle.

Insights into phosphatase-activated chemical defense in a marine sponge holobiont

Takahiro Jomori et al.

Summary: Marine sponges often contain potent cytotoxic compounds that are detoxified by phosphorylation catalyzed by the phosphotransferase CalQ of a producer symbiont. The phosphatase CalL, specific to phosphocalyculin A, coordinates the heteronuclear metals Cu and Zn and is localized in the periplasmic space of the sponge symbiont for on-demand production of calyculin A in response to tissue disruption.
