Association of CYP3A5 polymorphisms and parathyroid hormone with blood level of tacrolimus in patients with end-stage renal disease

Ryota Tanaka et al.

Summary: The study found that CYP3A5 polymorphism and PTH levels are associated with the blood concentration/dose ratio of tacrolimus in patients undergoing kidney transplantation. CYP3A5 expressors had lower tacrolimus concentrations, and there was a significant positive correlation between tacrolimus concentration/dose ratio and iPTH levels. Logistic regression analysis identified CYP3A5 polymorphism and serum iPTH level as significant factors associated with tacrolimus C/D ratio.

14,15-EET Reduced Brain Injury from Cerebral Ischemia and Reperfusion via Suppressing Neuronal Parthanatos

Haipeng Zhao et al.

Summary: The study found that 14,15-EET inhibits neuronal parthanatos induced by MCAO/R through upregulation of antioxidant gene expression and reduction of reactive oxygen species production. This advances the EET neuroprotection theory and provides a scientific basis for developing targeted clinical drugs to reduce neuronal damage following cerebral ischemia and reperfusion.

Soluble Epoxide Hydrolase Blockade after Stroke Onset Protects Normal but Not Diabetic Mice

Catherine M. Davis et al.

Summary: This study found that tAUCB administered after ischemic stroke onset provides brain protection in non-diabetic mice but not in diabetic mice, and the neuroprotection seems to be independent of changes in gross CBF. Furthermore, DM2-induced hyperglycemia abolishes t-AUCB-mediated neuroprotection after stroke onset.

Endothelium-Derived Hyperpolarizing Factor (EDHF) Mediates Acetylsalicylic Acid (Aspirin) Vasodilation of Pregnant Rat Mesenteric Arteries

Helga Helgadottir et al.

Summary: The study demonstrates that aspirin induces vasodilation in mesenteric resistance arteries by acting through the endothelial-derived hyperpolarizing factor (EDHF) and calcium-activated potassium channels. This mechanism suggests that aspirin may be beneficial for pregnancies complicated by hypertension by reducing peripheral vascular resistance.

Metabolism pathways of arachidonic acids: mechanisms and potential therapeutic targets

Bei Wang et al.

Summary: The arachidonic acid (AA) pathway is crucial in cardiovascular biology, carcinogenesis, and various inflammatory diseases, producing bioactive mediators that are potential targets for cardiovascular diseases, cancers, and inflammatory diseases. This review summarizes the physiological and pathophysiological importance of AA metabolizing pathways, as well as the molecular mechanisms underlying its actions in CVD and cancer progression. Additionally, clinical studies targeting AA metabolisms in different disease conditions are introduced to explore the role of AA metabolism and potential therapies.
