Practical aspects of preparation, optimization, and evaluation of poly (styrene-divinylbenzene)porous-layer open tubular (PLOT) columns for capillary liquid chromatography

Summary: Using open tubular (OT) capillary columns, specifically the PLOT (porous layer open tubular) type, offers advantages in terms of interaction with analytes in liquid chromatography. This study explores the application of PLOT columns in capillary LC and discusses the preparation, optimization, and evaluation of these columns. Poly(styrene-divinylbenzene) (PS-DVB) was used as the stationary phase, and comparisons were made between columns with internal diameters of 10 and 25 μm. The effects of flow rate and gradient window were also investigated.

Compact capillary high performance liquid chromatography system for pharmaceutical on-line reaction monitoring

Summary: Due to their size, conventional high performance liquid chromatographs (HPLCs) are difficult to place close to a reaction vessel within a pharmaceutical manufacturing or development site. However, a compact and modular separation system using capillary HPLC has been developed to enable co-location of an HPLC with a small-scale reactor for reaction monitoring. The compact system showed equivalent quantitative performance to an Agilent 1290 Infinity II HPLC system.

Porous layer open tubular nano liquid chromatography directly coupled to electron ionization mass spectrometry

Summary: A 25 μm i.d x 1.2 m length PS-DVB porous layer open tubular column (PLOT) was prepared and assessed in the configuration of a nano liquid chromatography coupled to an electron ionization mass spectrometry system (OT-nanoLC-EI-Ms). The system demonstrated competitive applicability in separating non-amenable ESI compounds, such as polyaromatic hydrocarbons (PAHs) and non-amenable GC compounds such as thermolabile pesticides. With excellent chromatographic performance, the PLOT columns can work under more compatible EI-detection conditions.

Summary: This study described the chromatographic and electrochromatographic separation of small neutral and charged analytes using a fused silica capillary with a stationary pseudophase semi-permanent coating of DDAB aggregates. The research showed the formation of DDAB patchy admicelle, complete admicelle, or larger aggregates inside the capillary, depending on the concentration of DDAB used in coating. Overall, the use of a stationary pseudophase coating provides a potentially easier alternative way to conduct open-tubular liquid chromatography and electrochromatography.
