Digital Microfluidics and Magnetic Bead-Based Intact Proteoform Elution for Quantitative Top-down Nanoproteomics of Single C. elegans Nematodes

Summary: Researchers have developed a sample preparation method based on protein aggregation and acidic elution, allowing for sensitive protein identification analysis of single nematodes on a digital microfluidics device. Compared to in-tube sample preparation, this method increases proteoform identifications by 46%. Antifouling Properties of Pluronic and Tetronic Surfactants in Digital Microfluidics

Summary: Fouling at liquid-solid interfaces is a severe problem for digital microfluidics (DMF) and can be combatted by including amphiphilic surfactant additives in the droplets. This study evaluated 19 different Pluronic and Tetronic additives to determine their antifouling performance, structural similarities, and mechanism of action. The analysis showed that Pluronic and Tetronic additives with specific characteristics can provide exceptional antifouling performance in DMF. The results and their implications are important progress for the DMF community and can guide the selection of surfactants for manipulating biological matrices.

All-in-One digital microfluidics pipeline for proteomic sample preparation and analysis

The pipeline allows for automated sample reduction, alkylation, digestion, isotopic labeling, and analysis. The All-in-One pipeline shows potential for evaluating mass-limited samples and could be useful for proteomic cancer sub-typing.

The method utilizes an omniphobic substrate patterned with hydrophilic surface energy traps (SETS), which, when wet with a solvent, form a dense microdroplet array for local extraction of analytes from the tissue surface. The extracted analytes can then be analyzed using matrix-assisted laser desorption/ionization-mass spectrometry (MALDI-IMS) or ambient ionization-MS techniques. The PSSE method shows potential to increase the speed of ambient MS tissue imaging techniques by reducing the steps required for sample preparation.

This detergent enables facile droplet movement and is compatible with organic solvents, allowing for efficient isobaric tandem mass tag (TMT) labeling.
