Age-Related Increase in Lactate Dehydrogenase Activity in Skeletal Muscle Reduces Life Span in Drosophila

Liam C. Hunt et al.

Summary: The study found that skeletal muscle aging is characterized by increased glycolysis but reduced expression of glycolytic enzymes. Lactate dehydrogenase (LDH), an enzyme necessary for anaerobic glycolysis, was found to increase with aging and contribute to mortality. These findings indicate an important role for muscle LDH and glycolysis in aging.

Nicotinamide Mononucleotide Administration Amends Protein Acetylome of Aged Mouse Liver

Chengting Luo et al.

Summary: It has been reported that the activities of NAD(+)-dependent deacetylase decrease in the aging mouse liver, while NMN treatment can increase healthspans. However, the changes in the acetylomic landscape and the effects of NMN treatment on protein acetylation in the aging liver have not been studied. In this study, immunoprecipitation coupled with label-free quantitative LC-MS/MS was used to identify the acetylome and investigate the effects of aging and NMN on liver protein acetylation. The results showed that the acetylation levels of proteins involved in oxidation-reduction and metabolic processes increased in aged livers, while the acetylation levels of proteins involved in transport and translation processes decreased. NMN treatment inhibited the age-related increase in acetylation of proteins regulating fatty acid beta oxidation, the TCA cycle and valine degradation.

Quantitative Acetylomics Reveals Dynamics of Protein Lysine Acetylation in Mouse Livers During Aging and Upon the Treatment of Nicotinamide Mononucleotide

Jingshu Li et al.

Summary: Lysine acetylation is an important post-translational modification involved in regulating various cellular processes, including aging. However, the acetylome-wide analysis in mammalian tissues during aging is limited. This study combines antibody enrichment and label-free quantitative acetylomic analysis to investigate the changes in protein acetylation during natural aging and upon the treatment of NMN. The results identify previously unknown acetylation sites and highlight the potential of protein acetylation events as aging biomarkers. Furthermore, the study demonstrates the beneficial effects of NMN on age-related protein acetylation. These findings lay a critical foundation for understanding the functional role of protein post-translational modifications in healthy aging and disease conditions.

Summary: GCN5 and PCAF are lysine acetyltransferase enzymes that play important roles in histone acetylation and non-histone substrate acetylation in eukaryotes. This review focuses on the current understanding of non-histone acetylation by GCN5 and PCAF in budding yeast and mammalian systems, highlighting the diverse impact on cell physiology. Critical caveats and open questions are defined at the end of the review for all models.
