Modulatory role of copper on hIAPP aggregation and toxicity in presence of insulin

Summary: Aggregation of hIAPP is associated with beta-cell death in T2DM. pH, insulin, and metal ions are key factors in controlling hIAPP aggregation. Insulin inhibits hIAPP fibrillation by preventing dimerization, while zinc and copper have inhibitory effects with copper promoting toxic oligomers. Copper induces structurally distinct toxic oligomers of hIAPP in the presence and/or absence of insulin. These findings contribute to our understanding of beta-cell factors in hIAPP aggregation.

Challenges in characterizing the aggregation process have led to the development of various biophysical and chemical approaches, with studies suggesting that restoring hIAPP proteostasis could be a promising strategy for therapy.
