First detection/quantification of roxadustat in hair with a new liquid chromatography with tandem mass spectrometry method: Application to a treated patient

Summary: This study aimed to develop a LC-MS/MS method for measuring the concentration of roxadustat in hair and apply it to a patient undergoing chronic treatment. After decontamination, testosterone-D3 and phosphate buffer were added to 20 mg of hair, and the mixture was incubated and extracted for analysis. The method was accurate and precise, and successfully used to measure roxadustat in a treated patient.

In recent years, MALDI coupled with IMS has emerged as an attractive option for hair analysis, providing high spatial resolution and competing with conventional methods. This article provides a comprehensive overview of MALDI techniques and their applications in understanding hair analysis.

Segmental analysis and long-term monitoring of vadadustat in equine hair for the purpose of doping control

Summary: In drug metabolism studies in horses, non-targeted analysis by liquid chromatography coupled with high-resolution mass spectrometry can be used to rapidly identify potential biomarkers. The presence of interfering components is a common problem, but this study demonstrates that using intelligently generated exclusion lists can expand the coverage of analysis and improve identification of drug metabolites. The approach was successfully applied to identify 21 metabolites of Daprodustat and establish its metabolic pathway.

Pharmacokinetic Study of Vadadustat and High-Resolution Mass Spectrometric Characterization of its Novel Metabolites in Equines for the Purpose of Doping Control

The results showed that vadadustat was mainly excreted in urine as the parent drug along with some minor conjugated metabolites. The findings provide evidence for effective control of the misuse or abuse of vadadustat in horses.

Following IV administration, daprodustat exhibited low plasma clearance and volume of distribution, indicating low tissue distribution.
