Renal function estimating equations performance during pregnancy and postpartum

Dina E. Zaghloul et al.

Summary: The study aimed to evaluate the performance of renal function estimating equations compared to measured creatinine clearance (CrCl) during pregnancy and postpartum, and to assess which body weight provides the best performance. The results showed that CG (PPW) and CG (ABW) had the best performance during pregnancy and postpartum, respectively. Based on overall performance, CG (PPW) was the best during pregnancy, and 100/SCr was the best postpartum.

Interplay of Breast Cancer Resistance Protein (Bcrp/Abcg2), Sex, and Fed State in Oral Pharmacokinetic Variability of Furosemide in Rats

Sheena Sharma et al.

Summary: The poor and variable oral bioavailability of furosemide (FUR) is a critical challenge in pharmacotherapy. This study investigated the interplay of breast cancer resistance protein (Bcrp)-mediated transport, sex, and fed state on FUR pharmacokinetics (PK) in rats. The results suggest that the PK of Bcrp and/or Oat substrates could be sex-dependent in rats and that species and sex differences in Bcrp and Oat abundance in the kidney should be considered when scaling rat PK and toxicological data of Bcrp substrates. Furthermore, Bcrp inhibition should be evaluated in prospective clinical studies to assess its effect on FUR PK.

State of the Art and Uses for the Biopharmaceutics Drug Disposition Classification System (BDDCS): New Additions, Revisions, and Citation References

Giovanni Bocci et al.

Summary: The Biopharmaceutics Drug Disposition Classification system (BDDCS) is a classification system based on water solubility and metabolism/permeability rate, which can predict the role of enzymes and transporters in drug disposition. This article presents the application of BDDCS in 1475 drugs and active metabolites, including revisions of previous classification studies. It also discusses the uses of BDDCS in drug development.

Impact of Albumin Binding Function on Pharmacokinetics and Pharmacodynamics of Furosemide

Gerd Klinkmann et al.

Summary: This study investigated the efficacy of furosemide in relation to albumin binding and function. The results showed that albumin binding capacity had an impact on the efficacy of furosemide, with higher ABiC associated with lower free furosemide fraction and higher urinary furosemide concentration.

Pregnancy-Related Hormones Increase UGT1A1-Mediated Labetalol Metabolism in Human Hepatocytes

Raju Khatri et al.

Summary: Pregnancy-related hormones have been shown to influence the expression and function of UGT proteins, particularly enhancing UGT1A1-mediated metabolism of labetalol in human hepatocytes. This findings provide mechanistic insight into the increased clearance of labetalol observed in pregnant individuals.
