Carbene-Catalyzed Activation of 2-Aminobenzaldehyde for Access to Chiral Fluorescent Quinazolinone

Tiantian Li et al.

Summary: In this study, a carbene-catalyzed reaction was used to synthesize a chiral quinazolinone with a new activation mode of an aniline-like N-H moiety. The addition of the nitrogen atom of diphenyl o-aminobenzaldehydes via NHC activation to imines resulted in chiral quinazolinones with high yields and optical purities. The acidity of the N-H moiety was greatly increased through the formation of an acyl azolium intermediate, which was investigated using DFT calculations. Furthermore, the chiral quinazolinones displayed high fluorescence quantum efficiency.

Asymmetric Synthesis of Imidazo[2,1-a]isoquinolin-3-ones with Dihydroisoquinolines and N-substituted Amino Acids

Xiao-Hui Chen et al.

Summary: A mild direct synthesis of chiral imidazo[2,1-a]isoquinolin-3-one derivatives with dihydroisoquinolines and N-substituted amino acids has been developed. The use of easily accessible DCC as condensation reagent at ambient temperature allows for the assembly of a series of imidazo[2,1-a]isoquinolin-3-ones in low to good yields with low to excellent diastereomeric ratios. Interestingly, the use of N-substituted L-Threonine and L-Serine affords oxazino[2,3-a]isoquinolin-4-one derivatives with good diastereoselectivities.

Chiral polycyclic benzosultams from photocatalytic diastereo- and enantioselective benzylic C-H functionalization

Shaonan Zhang et al.

Summary: Asymmetric C(sp(3))-H functionalization is a useful method for introducing functionality and chirality onto hydrocarbon units. However, achieving stereodiscrimination between a strong C(sp(3))-H bond and a prochiral substrate, leading to the formation of vicinal stereogenic centers in one step, remains challenging. In this study, a photocatalytic diastereo- and enantioselective C(sp(3))-H functionalization/intramolecular cyclization reaction is reported, which produces biologically and synthetically important polycyclic benzosultams and fused tetrahydroisoquinolines with high stereocontrol.
