PTABS: A Unique Water-Soluble π-Acceptor Caged Phosphine

Summary: Caged phosphines with unique structures have been developed into efficient catalytic systems with advantageous properties. Our research group recently introduced a highly water-soluble caged phosphine: PTABS (KapdiPhos), and explored its applicability in cross-coupling reactions of biologically relevant halonucleosides and chloroheteroarenes. This account details our journey from ideation to the commercialization of these ligands.

The Road Less Traveled: Unconventional Site Selectivity in Palladium-Catalyzed Cross-Couplings of Dihalogenated N-Heteroarenes

Summary: The regiochemical outcomes of Pd-catalyzed cross-coupling reactions for most dihalogenated N-heteroarenes are well-established, but changes to the catalyst can lead to unconventional product due to different site selectivity. This Perspective surveys unusual cases controlled by ligands or catalyst speciation where divergent selectivity occurs. Some cases have established the mechanistic origin of inverted selectivity, while others remain unknown. The remaining challenges and opportunities in the field include understanding oxidative addition mechanisms, catalyst speciation, ring substituents influence, inverted selectivity for stubborn substrates, and minimizing over-reaction of di- and polyhalogenated substrates.

Summary: Phosphines in combination with transition metals have been crucial in catalytic processes. Caged phosphines offer control over steric and electronic properties, with research groups like Verkade, Gonzalvi and Stradiotto demonstrating their versatility. Our research group has also made significant progress in developing 1,3,5-triaza-7-phosphaadamantane-based caged ligands for modifying biologically useful nucleosides and heteroarenes.

Room-Temperature Amination of Chloroheteroarenes in Water by a Recyclable Copper(II)-Phosphaadamantanium Sulfonate System

Summary: This study discusses an efficient copper-based catalytic system for aminating chloroheteroarenes at ambient temperature in water, which can couple a wide variety of chloroheteroarenes with primary and secondary amines. The catalytic system not only promotes late-stage functionalization of active pharmaceutical ingredients, but also exhibits high efficiency at a very low concentration and can be recycled multiple times with minimal loss in activity. The pi-acceptor ability of the ligand PTABS is identified as a key factor for the high reactivity of the catalytic system, as confirmed by theoretical calculations.

Buchwald-Hartwig reaction: an update
