Biomimetic Nanoerythrosome-Coated Aptamer-DNA Tetrahedron/Maytansine Conjugates: pH-Responsive and Targeted Cytotoxicity for HER2-Positive Breast Cancer

Wenjuan Ma et al.

Summary: DNA materials have been used as nanocarriers for targeted cancer therapy, but their short half-life and low bioavailability hinder their clinical translation. This study developed a HER2-targeted DNA-aptamer-modified DNA tetrahedron/DM1 conjugate, and applied a biomimetic camouflage to enhance drug delivery and tumor-stimulated drug release. The hybrid erythrosome-based nanoparticles exhibited superior inhibition of HER2-positive cancer.

Epidermal growth factor/epidermal growth factor receptor signaling blockage inhibits tumor cell-derived exosome uptake by oral squamous cell carcinoma through macropinocytosis

Eri Sasabe et al.

Summary: The study found that treatment with epidermal growth factor promoted the uptake of oral squamous cell carcinoma cell-derived exosomes into OSCC cells, while EGFR inhibitors could inhibit this process, suggesting that EGFR inhibitors may inhibit the malignant potential of OSCC cells by blocking the uptake of exosomes.

Gamma delta (?d) T cells in cancer immunotherapy; where it comes from, where it will go?

Jiechu Deng et al.

Summary: Cancer immunotherapy is an important approach in the treatment of cancer, and gamma delta T cells have shown great potential in this field. Gamma delta T cells can recognize a wide range of antigens, exert anti-tumor effects, and can be manipulated for therapeutic interventions. However, research on gamma delta T cells is still in its infancy and faces many challenges and limitations.

Current status and perspective of tumor immunotherapy for head and neck squamous cell carcinoma

Chenhang Yu et al.

Summary: Head and neck squamous cell carcinoma (HNSCC) has a high incidence and mortality rate. Immunotherapy, with its potential for a broad and long-lasting antitumor response, has become an important therapeutic strategy for HNSCC. However, immune escape limits the effectiveness of immunotherapy. Various immunotherapy strategies, including immune checkpoint inhibitors, costimulatory agonists, antigenic vaccines, oncolytic virus therapy, adoptive T cell transfer, and targeted therapy, have been developed for HNSCC. Each strategy has its unique advantages, and their appropriate application has significant value for the treatment of HNSCC.

TRAP1 suppresses oral squamous cell carcinoma progression by reducing oxidative phosphorylation metabolism of Cancer-associated fibroblasts

Li Xiao et al.

Summary: This study found that CAFs in oral squamous cell carcinoma exhibit greater mitochondrial activity and preferentially use oxidative phosphorylation for glucose metabolism. TRAP1 was identified as an important regulatory molecule in CAFs, promoting OSCC progression by regulating OXPHOS.

Exosome-derived microRNAs in oral squamous cell carcinomas impact disease prognosis

Ching-Mei Chen et al.

Summary: The study indicates that exosomal miR-155 and miR-21 are upregulated in OSCC patients, while miR-126 is downregulated. MiR-155 and miR-21 promote cell proliferation and invasion by suppressing PTEN and Bcl-6 tumor suppressors, while miR-126 inhibits oncogenic behaviors and EGFL7 expression. Exosomal miRNAs in blood may serve as biomarkers for the diagnosis and prognosis of OSCC.

miR-34a-5p functions as a tumor suppressor in head and neck squamous cell cancer progression by targeting Flotillin-2

Xiang Li et al.

Summary: The study demonstrates that downregulation of miR-34a-5p is associated with the progression of HNSCC tumors and cells. Functionally, miR-34a-5p inhibits proliferation, migratory/invasive activity, and epithelial-mesenchymal transition in HNSCC cells. Mechanistically, miR-34a-5p suppresses FLOT-2 expression and activates the MEK/ERK1/2 pathway.

Cancer-derived exosomal miR-138-5p modulates polarization of tumor-associated macrophages through inhibition of KDM6B

Jing Xun et al.

Summary: This study demonstrates that miR-138-5p delivered from breast cancer cells to tumor-associated macrophages via exosomes can downregulate KDM6B expression, inhibit M1 polarization, and stimulate M2 polarization, ultimately promoting tumor metastasis. Additionally, exosomal miR-138-5p levels positively correlate with the progression of breast cancer, suggesting it as a potential prognostic marker and therapeutic target.

OSCC cell-secreted exosomal CMTM6 induced M2-like macrophages polarization via ERK1/2 signaling pathway

Xin Pang et al.

Summary: In OSCC samples, higher expression of CMTM6 is associated with higher pathological stage, CD163+ macrophage infiltration, and PD-L1 expression. Knockdown of CMTM6 inhibits proliferative, migrative, and invasive abilities of OSCC cells, and suppresses M2 macrophage polarization in vitro by downregulating PD-L1 expression. Exosomal CMTM6 from OSCC cells promotes M2-like macrophage polarization via ERK1/2 signaling, indicating a novel crosstalk between cancer cells and immune cells in OSCC microenvironment.
