Concussion susceptibility is mediated by spreading depolarization-induced neurovascular dysfunction

Ellen Parker et al.

Summary: Parker et al. demonstrate that susceptibility to concussion is associated with blood-brain barrier dysfunction and neuroinflammation. Pharmacological treatment targeting the inflammation induced by blood-brain barrier dysfunction can protect brain function and improve neurological outcomes in a rodent model of traumatic brain injury. These findings are important for a better understanding of the complications of mild traumatic brain injury and the development of diagnostic and treatment methods.

Traumatic Brain Injury and Risk of Neurodegenerative Disorder

Benjamin L. Brett et al.

Summary: This article reviews the evidence for traumatic brain injury (TBI) as a risk factor for neurodegenerative disorders, including Alzheimer's dementia and Parkinson's disease, in clinical and neuropathological studies. It also describes the dynamic interactions between acute response to injury and chronic processes that may be involved in TBI-related pathogenesis and progression of neurodegeneration.

Quantitative Water Permeability Mapping of Blood-Brain-Barrier Dysfunction in Aging

Jeremy N. Ford et al.

Summary: Blood-brain-barrier (BBB) dysfunction is a key feature of aging and aging-related disorders. A study found that BBB water exchange rate (k(W)) differs in various brain regions, with higher values in the cerebral cortex compared to cerebral white matter. This difference is also observed in cerebral blood flow (CBF). Age was found to be statistically significant in relation to k(W) and CBF. Furthermore, there were moderate negative correlations between k(W) and white matter hyperintensity (WMH) volume. This research provides insights into the relationship between BBB water exchange and aging, and has implications for potential BBB-targeted therapies for aging-related brain disorders.

Ageing is associated with maladaptive immune response and worse outcome after traumatic brain injury

Federico Moro et al.

Summary: Traumatic brain injury is increasingly common in older individuals and is associated with poor prognosis. Aging is associated with extra-cranial comorbidities and pre-existent pathologies, as well as a dysregulated and chronic inflammatory immune response. Studies on mice and clinical cohorts have shown that aged individuals have greater functional deficits and worse long-term outcomes after brain trauma, which are related to altered immune response in systemic, meningeal, and brain tissue.

Modeling the Dynamics of a Secondary Neurodegenerative Injury Following a Mild Traumatic Brain Injury

Ryan M. Kochis et al.

Summary: GFAP is injected into the bloodstream during a traumatic brain injury, leading to increased IgG production to remove excess protein. Elevated and prolonged GFAP levels can be observed in repeated traumas, which may have implications for predicting brain lesions and understanding the recovery process post-TBI through simple blood tests.
