Preserved blood-brain barrier and neurovascular coupling in female 5xFAD model of Alzheimer's disease

Summary: Using two-photon microscopy and microscopy, this study observed the function of cerebral vasculature and neurovascular coupling in an AD mouse model. The results showed that the blood-brain barrier and neurovascular coupling were preserved, and the density of capillary pericytes was not affected. These findings suggest that microvascular function is preserved in the 5xFAD mice and highlight the importance of choosing appropriate preclinical models of AD.

Summary: The study demonstrated that atorvastatin could facilitate BBB maturation in remodeling vasculature in ischemic brain after stroke, reducing infarct size, restoring cerebral blood flow, improving neurologic outcomes. Atorvastatin promoted angiogenesis, stabilized newly formed vessels, and facilitated BBB properties in the new vessels by promoting endothelial tight junction formation. Additionally, pericytes expressing NG2 played a key role in mediating the effects of atorvastatin on BBB maturation.

This review sheds light on the critical role that glycocalyx plays in the function of the blood-brain barrier.

Reactive astrocytes facilitate vascular repair and remodeling after stroke

Summary: This study demonstrates that reactive astrocytes play a crucial role in vascular repair and remodeling after ischemic stroke in mice, by activating transcriptional programs related to vascular remodeling. Ablation of reactive astrocytes significantly impairs vascular and extracellular matrix remodeling, leading to prolonged blood flow deficits, exacerbated vascular permeability, ongoing cell death, and worsened motor recovery. These findings highlight the critical role of reactive astrocytes as cellular mediators of vascular remodeling during neural repair.

Chronic exposure to Aβ impairs EDN1-mediated pericyte contractility, potentially contributing to neurovascular uncoupling and reduced cerebral blood flow in AD.

Understanding the role of the glycocalyx in DCI could lead to improved predictive methods and treatment strategies.
