GSDMD knockdown exacerbates hippocampal damage and seizure susceptibility by crosstalk between pyroptosis and apoptosis in kainic acid-induced temporal lobe epilepsy

Summary: Neuronal loss is a vital feature of temporal lobe epilepsy, and the exact mechanism is not fully understood. This study investigated the involvement of gasdermin D (GSDMD)-mediated pyroptosis and the crosstalk among pyroptosis, apoptosis, and necroptosis in the pathogenesis of TLE. The results suggest that GSDMD-mediated pyroptosis contributes to TLE, but inhibition of GSDMD triggers crosstalk between pyroptosis and apoptosis, exacerbating neuronal loss and seizure susceptibility.

The disease involves the hyperphosphorylation and aggregation of Tau protein, the formation of Amyloid-β plaques, and neuroinflammation mediated by microglia. Microglia sense the release of aggregated Tau and migrate towards damaged neurons through purinergic P2Y12R signaling and actin remodeling.

By scavenging reactive oxygen species, inhibiting activation of the inflammasome, and reducing the production of inflammatory factors, PBzyme exerts neuroprotective effects.
