GSK343, an Inhibitor of Enhancer of Zeste Homolog 2, Reduces Glioblastoma Progression through Inflammatory Process Modulation: Focus on Canonical and Non-Canonical NF-κB/IκBα Pathways

Sarah Adriana Scuderi et al.

Summary: This study investigated the effect of GSK343 on canonical/non-canonical NF-kappa B/I kappa B alpha pathways and immune response in GB. In vitro results showed that GSK343 significantly reduced GB cell viability and modulated pathway activation. In vivo, GSK343 reduced tumor mass and regulated oxidative stress levels. Ex vivo results confirmed the anti-proliferative effect of GSK343 and demonstrated its ability to regulate immune response.

EZH2 inhibition decreases neuroblastoma proliferation and in vivo tumor growth

Laura Bownes et al.

Summary: The EZH2 inhibitor GSK343 has significant effects on the treatment of neuroblastoma, significantly reducing cell viability, migration, and stemness, while inhibiting tumor growth. This suggests that further research into the anti-tumor mechanisms of EZH2 inhibitors in neuroblastoma cells is warranted.

Histone Methylation Regulation in Neurodegenerative Disorders

Balapal S. Basavarajappa et al.

Summary: Advances in molecular biology and genomics have uncovered the critical role of epigenetic mechanisms, particularly histone methylation, in neurodegenerative disorders. Potential therapeutic targets, such as histone methyltransferases and demethylases, offer promising avenues for treatment and prevention. These findings bridge molecular-level changes with behavioral differences and present new opportunities for early intervention in individuals at risk for neurodegenerative disorders.

Summary: Glioblastoma is an aggressive brain malignancy with a poor prognosis, and the limited success of immunotherapy in treating it is mainly due to inadequate tumor infiltrating lymphocytes (TILs) and restricted systemic T cell trafficking into the central nervous system (CNS).
