Autophagy regulates inflammation in intracerebral hemorrhage: Enemy or friend?

Secondary brain injury (SBI) following ICH is mainly caused by inflammation, oxidative stress, and excitotoxicity. Haste makes waste: staged suppression of autophagic-lysosomal pathway in microglia promotes the efficient clearance of myelin debris

Summary: The autophagic-lysosomal pathway in microglia is crucial for removing myelin debris in white matter damage. However, the regulation of this pathway and maintaining lipid metabolism balance are still unclear. Excessive activation of autophagy causes lipid overload and accumulation in lysosomes and lipid droplets, leading to microglial dysfunction and secondary white matter damage. Suppression of autophagic activation in the acute phase of demyelination benefits microglia by restoring lipid metabolism balance and promoting myelin debris removal. Neuroprotective effects of microglial autophagy regulation may involve intracellular linoleic acid (LA) production and activation of the PPARG pathway.

Myeloid Trem2 Dynamically Regulates the Induction and Resolution of Hepatic Ischemia-Reperfusion Injury Inflammation

Summary: Trem2 plays a complex and heterogeneous role in liver inflammation, with its function changing at different stages. In the early stage of inflammation induction after hepatic ischemia-reperfusion (IR), myeloid Trem2 deletion inhibits the induction of inflammatory markers and reduces neutrophil accumulation and mitochondrial damage. However, as pro-inflammatory macrophages transition into resolution macrophages, Trem2 promotes phagocytosis of apoptotic cells, contributing to the resolution of inflammation. Overexpression of myeloid Trem2 accelerates the transition of monocytes to a resolution phenotype, while knockdown of Trem2 delays upregulation of certain markers. Overall, Trem2 is involved in the cascade of IR inflammation in a two-sided capacity.

NLRP3 inflammasome has been identified as a critical mediator of neuroinflammation, and inhibiting NLRP3 activation can alleviate AD-related pathology. Several synthetic and natural compounds have shown potential in inhibiting NLRP3 inflammasome and could be used as therapeutic interventions for AD.

Suppression of excessive autophagic-lysosomal activation can improve myelin debris degradation, while supplementation with linoleic acid can attenuate inflammation and restore microglial pro-regenerative properties, ultimately promoting recovery from demyelination injuries and improving spatial learning function.
