Summary: Amyloid-beta precursor protein (APP) regulates neuronal activity through the release of secreted APP (sAPP) acting at cell surface receptors. A 17 amino acid peptide (APP17) derived from APP binds to the extracellular sushi domain 1 (SD1) of GABA(B) receptors (GBRs). However, APP17 does not influence GBR activity in heterologous cells, indicating that sAPP exerts its neuronal effects through receptors other than GBRs.

Summary: The expression and functional role of BACE2 in the central nervous system (CNS) are still unclear. Previous studies have identified mutations in BACE2 in Hirschsprung disease (HSCR) patients, and this research confirms that these mutations are loss-of-function mutations. Using brain organoids, the study demonstrates that BACE2 is mainly expressed in certain areas and its expression increases with the maturation of the organoids. Functionally, brain organoids with the mutated BACE2 show phenotypes resembling Alzheimer's disease (AD), while overexpression of wild-type BACE2 attenuates AD-associated phenotypes.

Summary: This study used partial endothelial nitric oxide synthase (eNOS)-deficient mice as a model of age-dependent, spontaneous cerebral small-vessel disease (CSVD) and found that these mice exhibited cerebral hypoperfusion and blood-brain barrier leakage at a young age, with symptoms worsening progressively with age, ultimately leading to brain damage and cognitive impairment. The research also suggested that the bone morphogenetic protein 4 (BMP4) signaling pathway in brain pericytes may be a potential therapeutic target for age-related brain vascular diseases.
