Deepening the role of excitation/inhibition balance in human iPSCs-derived neuronal networks coupled to MEAs during long-term development

Alginate-Laminin Hydrogel Supports Long-Term Neuronal Activity in 3D Human Induced Pluripotent Stem Cell-Derived Neuronal Networks

Summary: This study establishes 3D models of neural cultures using human induced pluripotent stem cell-derived neural progenitor cells embedded in hydrogels, either pure alginate or alginate functionalized with laminin 111 (L111). The addition of L111 enhances neural migration, differentiation, and synaptogenesis. The neural networks formed in these hydrogels are electrically active for a long period of time, and L111 supplementation further increases electrical activity, network maturation, and synchronicity.

Long-term calcium imaging reveals functional development in hiPSC-derived cultures comparable to human but not rat primary cultures

Summary: Models for human brain-oriented research often rely on rodent primary cultures, which lack cellular specificity and molecular cues. In this study, we compared human induced pluripotent stem cell (hiPSC)-derived cultures with rat primary cultures for their dynamic and functional behavior. We found that hiPSC-derived cultures exhibited progressively richer activity patterns and functional traits during development, making them excellent models for studying neuronal assembly development and monitoring alterations caused by damage or neurodegeneration.

Human and mouse cortical astrocytes: a comparative view from development to morphological and functional characterization

Summary: The traditional view of astrocytes as a mere scaffold to neuronal circuitry has been debunked. Astrocytes not only have a neurotrophic function, but also actively participate in supporting synaptic transmission and calibrating blood circulation. Studies conducted in murine models have revealed many aspects of their functioning, but evidence shows significant differences between mouse and human astrocytes, including their development, morphology, transcriptomics, and physiology. The pursuit of superior cognitive abilities unique to humans has impacted the structure of the neocortex, affecting astrocytes along with the neuronal circuitry, resulting in species-specific properties. In this review, we summarize the diversities between murine and human astroglia, with a specific focus on neocortex, encompassing their developmental origin and all the structural and molecular differences that make human astrocytes unique.

Generation of glutamatergic/GABAergic neuronal co-cultures derived from human induced pluripotent stem cells for characterizing E/I balance in vitro

This approach allows for studying the cell-type-specific contribution of disease genes to E/I balance in human neurons.

Long-term morphological and functional dynamics of human stem cell-derived neuronal networks on high-density micro-electrode arrays

Summary: This study comprehensively characterized the development of human induced pluripotent stem cell (hiPSC)-derived neuronal networks using high-density micro-electrode arrays. The research revealed changes in morphology and activity features over time, including the formation of neuronal clusters and the transition to network-wide synchronized bursts. The study also found an increase in fast spiking neurons after 63 days post induction and identified the role of GABAergic neurotransmission in regulating spontaneous network bursts during circuit development and maturation.
