ERK Inhibition Promotes Engraftment of Allografts by Reprogramming T-Cell Metabolism

Summary: This study reveals the importance of the extracellular regulated protein kinases (ERK) signaling pathway in the development of allograft rejection. By using a specific ERK inhibitor called lycorine, the researchers found that inhibition of ERK significantly prolongs allograft survival and decreases the number and activation of infiltrating T cells. Further analysis shows that lycorine-treated T cells exhibit mitochondrial dysfunction and metabolic reprogramming, resulting in reduced responsiveness to stimulation. Transcriptome analysis also reveals downregulated immune response, mitogen-activated protein kinase cascade, and metabolic processes in lycorine-treated T cells. These findings provide new insights into the development of immunosuppressive agents targeting the ERK pathway involved in T-cell activation and allograft rejection.

Probing Indole Diketopiperazine-Based Hybrids as Environmental-Induced Products from Aspergillus sp. EGF 15-0-3

Summary: Comprehensive analysis of the metabolite spectra of Aspergillus sp. EGF 15-0-3 under different culture conditions revealed unique environment-induced metabolites exclusively found in the rice medium. Four unprecedented compounds were isolated, showing selective cytotoxicity against various human cancer cells. The putative biosynthetic pathways for these compounds involve Diels-Alder cycloadditions as key steps.

Summary: The study demonstrated that GPR30 agonist (G1) pretreatment or NLRP3 silencing significantly reduced the levels of inflammatory markers, improved histological changes and decreased hepatocyte apoptosis in a rat model of hepatic ischemia-reperfusion injury. These effects were associated with downregulation of NLRP3 expression and upregulation of GPR30 expression. GPR30 may serve as a potential therapeutic target for hepatic ischemia-reperfusion injury.

Indole Alkaloids from a Soil-Derived Clonostachys rosea

Bioassay-Guided Isolation of an Abetiane-Type Diterpenoid from Prunella vulgaris That Protects against Concanavalin A-Induced Autoimmune Hepatitis

Summary: Prunella vulgaris, a widely used edible Chinese medicinal plant, yielded two new abietane-type diterpenoids, abietoquinones A and B, which were found to possess immunosuppressive properties. Abietoquinone A demonstrated inhibitory effects on splenocyte proliferation and proinflammatory cytokine production, as well as protective effects on liver injury in Con A-challenged mice. Additionally, it showed inhibitory activities against the proliferation of murine splenocytes and human T cells induced by anti-CD3/anti-CD28 monoclonal antibodies (mAbs).
