Euphohelides A-C, ent-Abietane-Type Norditerpene Lactones from Euphorbia helioscopia and Their Anti-Inflammatory Activities

Summary: This study isolated three unreported ent-abietane-type norditerpene lactones, named euphohelides A-C (1-3), from the whole plants of Euphorbia helioscopia L. The structures of euphohelides A-C were established using spectroscopic methods, computational ECD, and X-ray crystallographic analyses. Euphohelide A (1) is a novel 2-nor-ent-abietane lactone with a unique 5/6/6/5 tetracyclic system.

Jatrophane Diterpenoids with Kv1.3 Ion Channel Inhibitory Effects from Euphorbia helioscopia

Summary: In this study, a total of 17 new jatrophane diterpenoids, named heliojatrone D (1) and helioscopids A-P (2-17), were isolated and identified from the whole plant of Euphorbia helioscopia. Additionally, 11 known analogues (18-28) were also found. The inhibitory effect of these compounds against Kv1.3 ion channels was evaluated, and two compounds (2 and 15) showed the most potent activity with IC50 values of 0.9 μM.

Ir-Catalyzed Biomimetic Photoisomerization of Cyclopropane in Lathyrane-Type Euphorbia Diterpenes

Summary: An efficient biomimetic method using iridium catalyst for the photoisomerization of cyclopropane in lathyrane-type Euphorbia diterpenes has been reported. The study successfully prepared lathyrane diterpenes with trans-fused cyclopropane using a stereochemical permutations strategy and verified the biogenesis relationship between different isomers. Additionally, the conversion from 5a to another trans-isomer, 5b, was also observed.

Bisfischoids A and B, dimeric ent-abietane-type diterpenoids with anti-inflammatory potential from Euphorbia fischeriana Steud.

Summary: Two undescribed ent-abietane-type diterpenoid dimers with nonacyclic backbone, bisfischoids A and B, were identified, along with a known compound fischdiabietane A, from Euphorbia fischeriana Steud. These compounds showed promising anti-inflammatory activities by inhibiting soluble epoxide hydrolase, with bisfischoid B exhibiting the strongest inhibitory effect. Further molecular docking studies revealed the key amino acid residue Tyr343 in the catalytic cavity of sEH for their inhibitory function.
