Sources of Antifungal Drugs

Giel Vanreppelen et al.

Summary: Due to their eukaryotic heritage, fungal pathogens have molecular similarities with their human hosts, making the development of novel antifungal drugs challenging. However, researchers have successfully identified effective candidates from natural or synthetic sources since the 1940s. These compounds have been enhanced and applied in clinical settings, offering valuable treatment for mycosis. Currently, there are only five different antifungal drug classes, leading to the development of antifungal resistance and a growing healthcare crisis.

Resolving a Natural Product Cold Case: Elucidation of Fusapyrone Structure and Absolute Configuration and Demonstration of Their Fungal Biofilm Disrupting Properties

Yun-Seo Kil et al.

Summary: Fusapyrones are fungal metabolites with broad-spectrum antibacterial and antifungal properties. This study successfully determined the full structures of fusapyrones and reinterpreted the absolute configurations of previously published metabolites using spectroscopic, chemical, and computational techniques. Biological testing demonstrated that fusapyrones inhibit and disrupt biofilm formation by Candida albicans, a human fungal pathoge.

Tackling the emerging threat of antifungal resistance to human health

Matthew C. Fisher et al.

Summary: This review emphasizes the important threat of invasive fungal infections to public health and the emergence of antifungal resistance. It discusses the main mechanisms of antifungal resistance and compares them with bacterial resistance to antimicrobial control. The review also explores the research and innovation topics needed to minimize the occurrence and impact of antifungal resistance, including environmental links, surveillance, diagnostics, transmission routes, novel therapeutics, and hotspot mitigation strategies. The need for global efforts to manage existing antifungal drugs and direct future research and development of therapies and interventions is highlighted.

Update on the Epidemiology, Diagnosis, and Treatment of Coccidioidomycosis

Samantha L. Williams et al.

Summary: Coccidioidomycosis is a fungal infection caused by Coccidioides immitis and Coccidioides posadasii, with increasing incidence and expanding geographic distribution. Most infections are asymptomatic, but some cases can be severe. Diagnosis is challenging and relies on clinician suspicion for laboratory testing. Advances in diagnostic tools and antifungal therapy aim to improve early detection and effective management of the infection.

Exploring the Promise of Endophytic Fungi: A Review of Novel Antimicrobial Compounds

Daniel J. Caruso et al.

Summary: The global emergence of antimicrobial resistance has rendered many existing drugs ineffective, leading to a demand for new, effective compounds. Endophytic fungi have shown great potential in producing novel bioactive compounds with broad antimicrobial activities. However, research in this field is still in its early stages.

Building Natural Product Libraries Using Quantitative Clade-Based and Chemical Clustering Strategies

Victoria M. Anderson et al.

Summary: This study proposes a set of tools combining genetic barcoding and metabolomics to help researchers build natural product libraries. Through experimentation with fungi, it was found that a modest number of isolates could provide a significant portion of chemical features, with different subclades containing varying levels of chemical diversity.
